Section Mécanismes de la Cancérogenèse (mca) Chef Dr Pierre Hainaut L’objectif général de la Section est de contribuer à la prévention et à la lutte contre le cancer à travers une meilleure compréhension des mécanismes de la cancérogenèse. Cet objectif implique l’étude des interactions entre l’environnement, le génome et l’épigénome. Une grande partie des activités de la Section consiste à réaliser des études translationnelles sur des biomarqueurs d’effet des expositions environnementales et des biomarqueurs de détection du cancer à un stade précoce, en concentrant les efforts sur les cancers les plus fréquents dans les pays à faible revenu, comme le carcinome hépatocellulaire (CHC), le carcinome épidermoïde des voies aéro-digestives et le cancer du sein. Les points forts des activités de la Section au cours de cette période biennale comprennent 1) le développement de techniques et de procédés pour permettre l’application des analyses de mutations multi-locus et de changements épigénétiques à de vastes études d’épidémiologie moléculaire et de pathologie moléculaire ; 2) de nouvelles lignes de recherche mécanistique concernant la contribution des mutations TP53 au développement de cancers spécifiques (poumon, sein, foie) et les bases moléculaires de la régulation épigénétique des cellules souches, en utilisant des systèmes élaborés de culture cellulaire in vitro ; 3) la mise en place et la coordination d’un Consortium international sur le cancer du foie (International Liver Cancer Study, http://ilcs.iarc.fr/); et 4) de nouvelles études sur la coordination des bases de données moléculaires, notamment de nouveaux développements pour la base de données des mutations TP53 du CIRC (http://www-p53.iarc.fr) et la mise en place d’un projet pilote pour la création d’une base internationale de données épigénétiques du cancer. La Section s’est également chargée de l’établissement et de la gestion d’une importante biobanque au CIRC, qui a gagné une notoriété internationale, en particulier grâce à la publication de directives et protocoles standard (Common Minimum Technical Standards and Protocols for Biological Resource Centres dedicated to Cancer Research), aujourd’hui reconnues comme référence mondiale pour les biobanques. section mécanismes de la cancérogenèse 39
