Introduzione U na grande varietà di sostanze chimiche presenti negli alimenti e nell’ambiente sono riportate avere attività sul sistema endocrino con meccanismi, bersagli <strong>ed</strong> effetti molteplici. gli interferenti endocrini (IE), sono un gruppo eterogeneo di composti utilizzati nella filiera agrozootecnica, in prodotti di consumo o industriali, accomunati dalla capacità di interferire a vari livelli del metabolismo e dell’azione degli ormoni, soprattutto steroidi sessuali e tiroi<strong>dei</strong>, causando effetti avversi su un individuo o sulla sua progenie durante lo sviluppo pre e post natale, spesso con effetti estrogenici o antiendrogenici. <strong>La</strong> fonte principale di esposizione nell’età evolutiva agli interferenti endocrini è rappresentata dall’alimentazione, in particolare durante gravidanza e allattamento1,4 , ma effetti endocrini possono essere esercitati anche da numerose sostanze presenti in cosmetici, filtri uV e altri prodotti per uso personale, giocattoli, materiali plastici, colle, dispositivi biom<strong>ed</strong>icali utilizzati in unità di cura neonatale intensiva 5-7 , evidenziando la ubiquitarietà di tali sostanze con una conseguente, potenziale esposizione multipla (ambiente di vita, ambiente scolastico, alimenti). Non sono disponibili, attualmente, informazioni sufficienti a stabilire in modo accurato e certo i possibili effetti a lungo termine legati alla esposizione cronica contemporanea alle basse concentrazioni ambientali di queste so- stanze che possono agire anche in modo additivo o sinergico. Inoltre, molti alimenti, anche di largo consumo, sono ricchi di interferenti endocrini di origine naturale (fitoestrogeni e micotossine) che potrebbero agire in sinergia o antagonismo con i contaminanti chimici ad azione ormono-simile 8,9 . ¢ Obiettivi A ttività di screening di primo livello su matrici alimentari e ambientali per evidenziare il rischio di esposizione della popolazione infantile ad interferenti endocrini. Vengono mostrati i risultati di prove sperimentali relative all’applicazione di un test a breve termine in vitro che misura la capacità di molecole o miscele ambientali complesse di riconoscere il recettore umano alfa per gli estrogeni, legarsi ad esso <strong>ed</strong> indurre una risposta biologica (attività estrogenica) nell’organismo test. Sono stati analizzati campioni di vari alimenti (alimenti ittici, acque da bere, principi attivi contenuti in prodotti vegetali <strong>ed</strong> erboristici) e materiali plastici utilizzati nel packaging alimentare e in dispositivi biom<strong>ed</strong>icali, per un totale di 133 campioni. ¢ Metodi P er lo studio è stata utilizzata una linea ricombinante di lievito S. cerevisiae transfettata con il recettore umano alfa per gli estrogeni (hEra).
