were sterilized and have anthers removed. By using different physical and chemical stress factors,<br />
androgenesis were induced in immature and unfertilized anthers. After putting anthers into Petri<br />
dishes, with growth media, cell dividing occurred. Anthers then developed callus tissues. Growth<br />
media have an impact on callus, allowing somatic embryogenesis to take place. During this<br />
process, new haploid plant was formed. Calli and regenerants were obtained. Data statistic<br />
analysis showed differences between abilities of barley genotypes to create calli and embryous.<br />
The creation of double haploid barley populations by anther technique can be used in barley<br />
breeding programmes.<br />

OBSERVATION OF SOMACLONAL VARIATION IN RED CLOVER<br />
(TRIFOLIUM PRATENSE L.) IN VITRO<br />

Red clover have been used to investigate plant regeneration and the occurrence of somaclonal<br />
variation. Somatic embryogenesis in vitro has been reported to occur either directly from<br />
predetermined embryogenic cells, such as embryonic or nuclear cells, or indirectly from<br />
differentiated cells which have dedifferentiated and subsequently been redetermined for<br />
embryogenic development. For obtaining of calli and somatical embyous, red clover variety ‗Agra‘<br />
was used. Was obtained viable regenerants from embryos. Somaclonal variation can be traced at<br />
the molecular level, evaluating the subtle adjustment of nuclear DNA. The high degree of diversity<br />
somaclones depends on the initial genotype, nature and stage of the explant. Was used flow<br />
cytometry analysis. Regeneration better get on top callus tissues, there is variation in ploidy. DNA<br />
was extracted from calli in acceptable quality and quantity. Extracted samples were frozen at -<br />
20Cº, and are available for further study using RAPD.<br />

RESULTS OF FLOW CYTOMETRY OF RED CLOVER (TRIFOLIUM<br />
PRATENSE L.) WILD POPULATIONS<br />

Red clover (Trifolium pratense L.) is an important forage legume widely cultivated in most<br />
temperate regions because of its characteristics of high seedling vigor, rapid growth, and tolerance<br />
to acidic and humid conditions. Red clover has a diploid genome (2n = 2x = 14), with a DNA<br />
content of 0.97 pg/2C. A diploid can have great flower substance, although if the same diploid<br />
plant is converted to a tetraploid, the plants characteristics will be enhanced. Previously efficient<br />
samples was filtered through a Partec 50 mkm CellTrics disposable filter and analysed in Partec<br />
Flow Cytometer using UV-laser and measured blue emission. Fifty Trifolium pratense L. samples<br />
from the Herbarium of Daugavpils University Institute of Systematic Biology were used.<br />
Approximately 200 cells were measured with a minimum of double analyses performed for each<br />
sample. All samples tested from different <strong>par</strong>ts of plants were similar and corresponded diploid<br />
level.
