QUALITATIVE DNA EXTRACTION FROM A DRY COLLECTION OF
MATERIAL FROM ARPEDIUM AND EUCNECOSUM GENUS
Key words: Arpedium and Eucnecosum genus, DNA extraction
Arpedium and Eucnecosum genus beetles sizes range from 1 to 35 mm, with most in the 2-8 mm
range. The form is generally elongate, with some being ovoid in shape. Colours range from yellow
to reddish-brown to brown to black. It is prevalent on every continent except Antarctica. Due to
the lack large-scale research of group systematics, the relationship between families and genus are
poorly researched. There is also no data about phylogenetic research of genus. For DNA extraction
was used a dry collection of beetles. Qualitative DNA extraction is difficult due to its
fragmentation. At the same time, DNA outcome is often a small sample of small size. For
qualitative DNA extraction was used MACHEREY-NAGEL protocol with modifications. As a
result, isolated DNA observed the corresponding quantity and quality. Specimens will be used for
future CO1, 18S and 28S gene amplification and sequencing.
THE STUDY OF MORPHOMETRY OF ADULT EUROPEAN POND
TURTLES EMYS ORBICULARIS IN LATVIA
Key words: Emys orbicularis, morphometry, Latvia
Adult European Pond Turtles Emys orbicularis found in Latvia (n=17) Weight (g) and standard
linear measuring CL, CB, Pl, PH, PB2, NuL, NuB (Meeske 2006: original U.Fritz) of carapax and
plastron (mm) were analyzed. Adults Emys orbicularis CL=161±17 mm, CB=126±13 mm. Adults
Emys orbicularis Weight=793±246 g. Correlation exists between CL and Weight, as well as
between CB and Weight. The values of the measured indications strongly correlate with each
other. The significant sexual differences of Emys orbicularis male and female adults exist for the
values of indications PBl, PB2, PH, Weight. There are no correlations of the values NuL and NuB
with the values of other indications. All values of linear sizes and Weight, excluding NuL and NuB
of Emys orbicularis male and female adults, correlate significantly with each other.
