CHAPTER 2: Introduction<br />

lead to pain and limitation <strong>of</strong> joint mobility, which decrease the quality <strong>of</strong> life for the patients and<br />

become a burden on the society 5 . Today, treatment <strong>of</strong> OA is based on anti-inflammatory drugs<br />

(e.g. NSAID), analgesics or arthroplastic surgery 12 , which neither slow down nor cure the disease.<br />

Moreover, no disease modifying osteoarthritic drugs (DMOADS) have been developed and<br />

approved by the American Food and Drug Administration (FDA) yet. Therefore, the discovery<br />

and the development <strong>of</strong> effective disease-modifying drugs are urgently needed.<br />

The large number <strong>of</strong> people affected by OA is increasing<br />

very fast. The increased life expectancy <strong>of</strong> the elderly<br />

population and the obesity epidemic, affirms the necessity<br />

to find a treatment for OA as fast a possible: Both to<br />

prevent an “OA-epidemic”, but also to avoid the great<br />

social-economical costs in the future.<br />
