Lipophilicity assessment of spironolactone by means of reversed phase liquid... 241ABFigure 2. Exemplary TLC chromatogram of examined spironolactone obtained on chromatographic plates RP-18F 254 using methanol ñwater: 90 : 10 (v/v) (A); example of HPLC chromatogram of spironolactone investigated on column RP18 by the use of methanol - waterin volume composition: 90 : 10 (v/v) (B)5) gives the result of lipophilicity descriptor (designatedas logP 1 ) very similar to logP exp . Other partitioncoefficients calculated by means of varioustopological indices according to Eq. 6 and Eq. 7 presentedin Table 1 are logP 2 and logP 3 which showhigher similarity to logP average . It could be suggestedthat topological indices may be useful in predicationof lipophilic properties of steroid compounds like, for example, spironolactone. As it was accurately emphasized in introductionpart of this work, in order to estimate which ofthe theoretically determined (by different calculationsprocedures) partition coefficient may be a reliablemeasure of lipophilicity of examined biomoleculethere is a need to compare all calculated logPvalues with those obtained by appropriate experimentalmethod. Therefore, the third stage of thisstudy was the comparison and assessment of allobtained results. Compared experimental and calculatedlipophilicity descriptors for spironolactone arepresented in Figure 3.As results from Figure 3, of all chromatographicallydetermined lipophilicity descriptors(R MW ) the biggest similarity to logP exp shows R MWobtained on glass RP-HPTLC plates RP-18F 254 andRP-2F 254 developed with mobile phase: dioxane -water: RMWRP18 (d) and also RMWRP2 (d) . Amongcomputed partition coefficients, these which arecomparable to those are logP KOWWIN and xlogP 3. Theresults of RP-HPLC analysis indicate that the chro-
