Products Imaging Live Cells under Near-Native Conditions Leica Microsystems launches the Leica DFC9000, a monochrome microscope camera with a highly sensitive third-generation sCMOS sensor. The camera enables researchers to image live cells under near-native conditions, allowing them to gain a better understanding of cellular processes and dynamics. The sensor with its high quantum efficiency over the entire spectrum of light, provides a high signal-to-noise ratio to securely detect even faint signals. Compared to the second generation sensor, the maximum quantum efficiency increased by 14%, totaling up to 82% depending on wavelength. In combination with a very low noise level, this results in a crisp fluorescence signal against a dark background. The high sensitivity of the camera eliminates the need to monitor GFP-overexpressing specimens and protects cells from phototoxicity. The Company points out that their Neo 5.5 sCMOS camera is particularly useful regarding latest Light Sheet Microscopy developments: A particular micro fabrication technique used to produce a mirror array that both directs the excitation beam and holds the sample may revolutionize the field of 3D super-resolution microscopy, according to an international group of scientists. They demonstrated that their Single-Objective Selective- Plane Illumination Microscopy (soSPIM) allows researchers to examine the activity of single proteins or entire embryos on their existing microscope systems. To greatly improve handling, the silicon support frame has been shaped into a 3.05 mm compatible hexagon with micro-structured TrueGrip edges. One side of the hexagon includes a reference notch to for sample location during processing and loading. Chemistry has been further improved to produce stress-optimised films, resulting in robust and ultra-planar silicon nitride films. Manufacturing of the EM-Tec silicon nitride films include a proprietary cleaning process to deliver clean, debris free films. Available with several windows sizes and membrane thicknesses of 20, 50 and 200nm. Correlative Raman-SEM Imaging The WITec RISE microscopy mode for correlative Raman-SEM imaging is now compatible with the scanning electron microscope Zeiss Merlin. The integration of both techniques into one system greatly improves ease-of-use and accelerates the experimental workflow. It places both the objective and sample stage required for Raman microscopy within the SEM’s vacuum chamber. Thus the sample can remain under vacuum for both measurements and is simply transferred between the Raman and SEM measuring positions by a software-driven pushbutton mechanism using an extremely precise scan stage.
