2.4. Purification of the Immune-Defense <strong>Peptide</strong>s<br />

1. Solid-phase extraction (SPE) on prepackaged, disposable cartridges containing<br />

chromatographic reversed-phase (C18 being the most frequently used). Sorbent<br />

weight ranges from 0.1 to 10 g. This step can be performed either individually<br />

with a syringe or with a multiposition vacuum manifold or with specific 96-well<br />

plates.<br />

2. Disposable Ultrafree-CL (Millipore) ultrafiltration cartridge units (10 kDa<br />

nominal molecular weight cutoff).<br />

3. Methanol, acetonitrile, TFA are HPLC grade. Water is Ultrapure (MilliQ TM or of<br />

HPLC quality). A proposed system for elution, solution A: acidified water (0.05%<br />

TFA) and solution B: acetonitrile in acidified water.<br />

4. Reversed-phase high-performance liquid chromatography (RP-HPLC) columns<br />

(C8, C18) from semipreparative (7 mm internal ø) down to capillary (
