In fact, such kinds <strong>of</strong> difficulties are precisely why <strong>the</strong>Agency must look carefully at standards <strong>for</strong> PM that encompass <strong>the</strong> fullspectrum <strong>of</strong> potential effects in different locations.Chapter 7: Dosimetry <strong>of</strong> <strong>Particulate</strong> <strong>Matter</strong> - General Comments:Chapter 7 on <strong>the</strong> dosimetry <strong>of</strong> particulate matter primarily focuses on an update <strong>of</strong> new studies since <strong>the</strong>1996 <strong>Criteria</strong> <strong>Document</strong> (CD). While <strong>the</strong> chapter provides a reasonable review <strong>of</strong> <strong>the</strong> available literature,<strong>the</strong> review is lacking in details in a number <strong>of</strong> areas. Given <strong>the</strong> importance <strong>of</strong> and reference to dosimetryconsiderations elsewhere in <strong>the</strong> document, <strong>the</strong> chapter should be streng<strong>the</strong>ned with more specificpresentations <strong>of</strong> some <strong>of</strong> <strong>the</strong> latest results.The chapter fails to take advantage <strong>of</strong> a graphical representation <strong>of</strong> <strong>the</strong> more recent data. Such graphicalrepresentations covering susceptible subgroups in comparison to normal subjects would be <strong>of</strong> great value.The authors failed to report whe<strong>the</strong>r increases compared from one group to ano<strong>the</strong>r are actuallystatistically significant or just represent general trends. Without showing <strong>the</strong> data and <strong>the</strong> standarddeviations or error bars, <strong>the</strong> reader is left with a general uncertainty about <strong>the</strong> significance <strong>of</strong> anydifferences that are reported.Section 7.5 on <strong>the</strong> comparisons <strong>of</strong> deposition and clearance patterns <strong>of</strong> particles administered byinhalation intratracheal instillation adds little to <strong>the</strong> chapter. This section, while accurate, is <strong>of</strong> little value<strong>for</strong> <strong>the</strong> risk assessment <strong>of</strong> particulate matter. There is no mention <strong>of</strong> <strong>the</strong> role that intratrachealadministration can play in hazard identification and in mechanism <strong>of</strong> action studies. If this section isretained, clearer identification <strong>of</strong> <strong>the</strong> value <strong>of</strong> <strong>the</strong> animal toxicological studies using this method should bediscussed. This is particularly important since many <strong>of</strong> <strong>the</strong> studies presented in Chapter 8 on animaltoxicological results arise from intratracheal administration experiments. Section 7.5 should be reduced insize if it is retained.Detailed tables or graphs contrasting deposition in children compared to adults should be presented in <strong>the</strong>chapter. Since arguments are made elsewhere in <strong>the</strong> CD about children being a potential susceptiblepopulation, dosimetric differences between children and adults need to be presented in greater detail than<strong>the</strong>y currently are. The logic <strong>of</strong> having <strong>the</strong> only figure in <strong>the</strong> dosimetry chapter be one <strong>of</strong> total depositionis not apparent. While such data are <strong>of</strong> general interest, <strong>the</strong> types <strong>of</strong> effects and standard setting concernsfocus on <strong>the</strong> major regions <strong>of</strong> <strong>the</strong> respiratory tract. Regional deposition should be presented and shouldincorporate recent research on different subpopulations and disease groups.Specific Comments:p. 7-2, l. 16 The reference to in<strong>for</strong>mation related to <strong>the</strong> phenomenon <strong>of</strong> particle overload isstretching <strong>the</strong> case <strong>for</strong> inclusion <strong>of</strong> this material. Clearly, <strong>the</strong>re are no ambientexposures <strong>of</strong> particulate matter that approximate anything close to <strong>the</strong> exposurelevels needed to induce overload <strong>of</strong> alveolar macrophage-mediated clearancethat is <strong>the</strong> basis <strong>for</strong> this phenomenon in animals.p. 7-5, l. 7 11 The authors should clarify that <strong>the</strong> importance being described <strong>for</strong> variousdeposition mechanisms in respiratory tract regions applies to humans. Theimportance <strong>of</strong> some <strong>of</strong> <strong>the</strong>se mechanisms differs on a relative sense <strong>for</strong> someand on an absolute sense <strong>for</strong> o<strong>the</strong>rs when referring to particle deposition inanimals.p. 7-6, l. 21 27 The cast studies with charged particles are not very relevant to real worldambient aerosols. If this material is retained, a better explanation <strong>of</strong> where <strong>the</strong>seresults might be applicable <strong>for</strong> potential real world exposures should beprovided.p. 7-7, l. 6 16 It is important in this paragraph to contrast inhalability in humans compared toinhalability <strong>of</strong> particles in animals. O<strong>the</strong>rwise <strong>the</strong> reader has no insight as towhy this is an important concept to introduce and fur<strong>the</strong>r has no reference <strong>for</strong>determining some <strong>of</strong> <strong>the</strong> relevance <strong>of</strong> concentrations used in animals whenjudging <strong>the</strong> potential <strong>for</strong> effects in humans.p. 7-8, l. 1 7 Of value would have been to compare <strong>the</strong> recent results <strong>of</strong> Kim to thosepreviously published by <strong>the</strong> GSF group <strong>for</strong> various combinations <strong>of</strong> tidalvolumes and respiratory frequencies.p. 7-10, l. 14 19 Since <strong>the</strong> study by Lenin used a fairly narrow size range (0.3 2.5 mm), <strong>the</strong>A - 6
