Proof of long-term stored potato germplasm by use of molecular markers<br />

bands were counted for all genotypes. Identity of banding patterns between genebank<br />

accessions was detected for ten of the twelve cultivars (Tab. 1, Fig. 1). However,<br />

in the cultivars ‘Early Rose’ (Fig. 2) and ‘Epicure’, differences in banding patterns<br />

were found for five and ten SSR primer pairs, respectively (Tab. 1, Fig. 3). The<br />

following reasons are conceivable:<br />

• Inclusion of donor cultivars carrying genetic modifications at the start of the collection.<br />

• Occurrence of genetic changes during long-term storage caused by the use of<br />

growth retardants or environmental stress in handling of in vitro and in situ collections.<br />

• Instability of the cultivar.<br />

• Mistakes or mixing of cultivars during the activities of the conservation methods.<br />

Genotypes with different banding patterns should be further tested by comparison of<br />

morphological traits according to the description of the cultivar list. Material of the<br />

same cultivar should be ordered from a third genebank to compare the fingerprints.<br />

First results showed that DNA fingerprinting with SSR primers is a useful, rapid and<br />

reliable method to check cultivar identity in genebank collections. In further experiments<br />

additional cultivars and other types of germplasm like wild potato species will<br />

have to be included.<br />
