MODERATE INCREASE IN MATERNAL CORTICOSTERONE REDUCES WITHIN-GROUP VARIATION IN BEHAVIOURAL, ENDOCRINE AND IMMUNE PARAMETERS IN ADULT MOUSE OFFSPRING Simone Macrì and Giovanni Laviola * . Section of Behavioural Neuroscience, Department of Cell Biology & Neuroscience, Istituto Superiore di Sanità, Viale Regina Elena 299, 00161 Roma, Italy The significance of behavioural neuroscience and the validity of its animal models of human pathology largely depend on the possibility to replicate a given finding across different facilities. However, when several mouse strains are tested on highly standardised behavioural test batteries in different laboratories, significant strain x lab interactions can be readily observed (Crabbe et al., 1999; Wolfer et al., 2004). Withingroup variation explains most of the variation observed in behavioural data, thus pointing at inter-individual variation as the major source of limited experimental reproducibility (Wolfer et al., 2004). Here we show that moderate doses of neonatal corticosterone supplementation through maternal drinking water (low, L-CORT, 33µg/ml or high, H-CORT, 100µg/ml) during the first lactation week, induce a significant reduction in inter-individual variation in adult male offspring in pain response, novelty preference, corticosterone response and immune competence. Our data demonstrate that a pharmacological simulation of increased neonatal environmental demands (Catalani et al, 1993) reduces experimental noise in adult laboratory rodents. Additionally, in spite of its effects on inter-individual variation, early corticosterone treatment did not affect normal (Gaussian) data distribution. Quite the opposite, whereas control subjects’ frequency distribution showed a mild yet significant deviation from normality, both L-CORT and H-CORT adult offspring showed a normal distribution. These results will foster new studies aimed at modifying the neonatal environment such as to induce moderate levels of stimulation and in turn reduce experimental noise (Macrì and Wuerbel, 2006). Importantly, rather than being limited to a specific variable, reduced variation was observed in a number of parameters, thus suggesting that these results may apply to a relatively broad spectrum of behavioural and physiological systems. Primarily, this study will lead to a reconsideration of the basic principles of neonatal housing systems, and ultimately,
this effort should induce more stable experimental models of human disorders. Finally, these studies will lead to a substantial reduction in the number of mice used in laboratory experiments.
