BlackPearl-L (Wheat Germ Lectin)<br />
Wheat germ lectin (from Triticum vulgaris) conjugated<br />
to BlackPearl Magnetic Beads. Wheat germ agglutinin<br />
is a 36,000 molecular weight protein consisting<br />
of two identical subunits. The receptor sugar for WGA<br />
is N-acetylglucosamine, with preferential binding to<br />
dimers and trimers of this sugar. WGA can bind oligosaccharides<br />
containing terminal N-acetylglucosamine<br />
or chitobiose, structures which are <strong>com</strong>mon to many<br />
serum and membrane glycoproteins. Bacterial cell wall<br />
peptidoglycans, chitin, cartilage glycosaminoglycans<br />
and glycolipids can also bind WGA.<br />
• Inhibiting/Eluting Sugar: 500mM N-acetylglucosamine,<br />
Chitin Hydrolysate, or 100 mM acetic acid<br />

BlackPearl-A (Protein-A)<br />
An affinity matrix ideal for isolation and purification<br />
of immunoglobins on a small scale. Black<br />
Pearl-A magnetic beads consist of re<strong>com</strong>binant<br />
Protein A covalently coupled to BlackPearl.<br />
Protein A has a high affinity for subclasses of<br />
IgG from a variety of species, including human,<br />
rabbit, mouse and sheep. The matrix can be<br />
regenerated up to 6 times with minimal loss of<br />
binding capacity. It can be used to immunoprecipitate<br />
target proteins from crude cell lysates<br />
using a selected primary antibody. Specific antibodies<br />
can be covalently cross-linked to the Black<br />
Pearl-A surface to create a reusable immunoprecipitation<br />
bead, thus avoiding the coelution<br />
of antibody with the target antigen.<br />

BlackPearl-G (Protein-G)<br />
<strong>Bio</strong>World BlackPearl-G Magnetic beads are<br />
prepared by covalent immobilization of Protein-<br />
G (from Streptococcus sp.) by CNBr activation<br />
coupling. Protein G shows strong binding affinity<br />
to the Fc region of several classes of mammalian<br />
immunoglobulins. BlackPearl-G has<br />
been used as a reliable tool to purify monoclonal<br />
and polyclonal IgGs from ascites, serum and<br />
cell culture supernatants. It is an ideal choice for<br />
affinity purification and/or fractionation of IgG<br />
subclasses which bind poorly to Protein-A such<br />
as bovine, goat, rat, and sheep IgG. Supplied as<br />
suspension in 100 mM Tris·Cl, pH 8.0, 500 mM<br />
NaCl, 0.05 % NaN 3 and 0.02 % thimerosal.<br />
