Activation of Complement by the Coagulation System
Introduction: Both, the complement and the coagulation system belong to the "first line
of defence" and undergo massive activation early after severe injury. A dialogue
between the two cascades has often been proposed but the precise molecular pathways
of this interaction are still unclear. To elucidate the mechanisms involved, the effects of
various coagulation factors on generation of the complement activation products C3a
and C5a was investigated.
Methods: In vitro cleavage of C5 or C3 in presence or absence of various coagulation
factors (F) was determined in a time- and dose-dependent manner by ELISA and
Immunoblotting. Biological activity of the cleavage products was also assessed by C3adependent
mast cell (HMC-1)- and C5a-dependent neutrophil-chemotactic activity. Oneway
