enhanced the <strong>in</strong>fectivity <strong>of</strong> JFH-1 4-fold at 48 hrs post <strong>in</strong>fection and this<br />
<strong>in</strong>creased to 8-fold by 72 hrs (Figure 5-1A). At 72 hrs the <strong>in</strong>creased <strong>in</strong>fectivity<br />
was associated with an <strong>in</strong>crease <strong>in</strong> the size <strong>of</strong> <strong>in</strong>fected cell foci comparable to<br />
that seen <strong>in</strong> Figure 4-3 (data not shown), suggest<strong>in</strong>g <strong>in</strong>creased cell-cell<br />
transfer <strong>of</strong> <strong>in</strong>fection. In contrast, JFH-1 G451R showed 1.5-fold <strong>in</strong>crease <strong>in</strong><br />
<strong>in</strong>fectivity at both time po<strong>in</strong>ts with no change <strong>in</strong> focal size (Figure 5-1A).<br />
To further <strong>in</strong>vestigate JFH-1 G451R <strong>in</strong>teraction(s) with SR-<strong>BI</strong> we employed<br />
the neutralis<strong>in</strong>g rabbit anti-SR-<strong>BI</strong> serum characterised <strong>in</strong> Figure 4-6 (120,<br />
190). Huh-7.5 cells were pre-<strong>in</strong>cubated with the anti-SR-<strong>BI</strong> sera prior to<br />
challeng<strong>in</strong>g with JFH-1 wt or G451R. Whilst the <strong>in</strong>fectivity <strong>of</strong> both viruses was<br />
<strong>in</strong>hibited, JFH-1 G451R is less sensitive to neutralisation; a 1/100 dilution <strong>of</strong><br />
antiserum reduced JFH-1 G451R <strong>in</strong>fectivity by 20%, compared to 80% for wt<br />
virus (Figure 5-1B).<br />
It is widely reported that HDL enhances HCV <strong>in</strong>fection via an SR-<strong>BI</strong><br />
dependent mechanism requir<strong>in</strong>g the transfer <strong>of</strong> lipids from HDL (25, 80, 324).<br />
To ascerta<strong>in</strong> whether the altered relationship <strong>of</strong> G451R with SR-<strong>BI</strong> extends to<br />
lipoprote<strong>in</strong> enhancement, JFH-1 wt and G451R <strong>in</strong>fections were supplemented<br />
with 10µg/ml HDL. HDL promoted JFH-1 wt <strong>in</strong>fectivity 2-fold, consistent with<br />
previous reports, however G451R <strong>in</strong>fectivity was unaltered (Figure 5-1C).<br />
Taken together, these data suggest that cell culture adaptation reduces the<br />
requirement for SR-<strong>BI</strong> dur<strong>in</strong>g virus entry.
