REGULATION OF MAPK MEDIATED OP18/STATHMIN SIGNALING PATHWAY<br />

TRIGGERED BY <strong>EBV</strong> ENCODED LMP1 IN NASOPHARYNGEAL CARCINOMA CELLS<br />

Oralabstract:<br />

Op18/stathmin is a molecule proposed to relay integrating diverse cell signaling pathways, the regulation<br />

<strong>of</strong> Op18/stahmin signaling can cause crucial changes <strong>of</strong> cellular biological properties including cell<br />

growth and proliferation. Our studies focused on the regulation <strong>of</strong> Op18/stathmin signaling by LMP1 in<br />

nasopharyngeal carcinoma (NPC) cells, Our results showed that LMP1 didn’t affect the expression <strong>of</strong><br />

Op18/stathmin , but LMP1 promoted the phosphorylation <strong>of</strong> Op18/stathmin. LMP1 was able to regulate<br />

Op18/stathmin signaling through MAPK mediation, but the activity <strong>of</strong> MAPK induced by LMP1 was not<br />

constant at different phases, changed with the cell cycle progression. LMP1 up-regulated the<br />

phosphorylation <strong>of</strong> MAPK at G1/S. On the contrary, LMP1 negatively regulated the phosphorylation <strong>of</strong><br />

MAPK at G2/M phase. Inhibition <strong>of</strong> LMP1 expression attenuated the interaction <strong>of</strong> MAPK with<br />

Op18/stathmin and promoted microtubule depolymerization. All these revealed a new pathway through<br />

which LMP1 regulates Op18/stathmin signaling by MAPK mediation. The new signaling pathway on<br />

LMP1 which is closely associated with cell cycle progression not only perfects the LMP1 regulation<br />

network, but also provides new insights for elucidating the molecular mechanism <strong>of</strong> LMP1 leading to<br />

tumorigenesis.<br />
