THE INVESTIGATION OF INTERACTION BETWEEN NASAL NK/T-CELL LYMPHOMA AND<br />

THE INFLAMMATION CELL.<br />

Oralabstract:<br />

Background: Nasal NK/T-cell lymphoma (NNKTL) is the lymphoproliferative disorder with necrotic<br />

granulomatous legions in the central part <strong>of</strong> head and neck region such as nasal cavity and hard palate. It<br />

is known Epstein–Barr virus (<strong>EBV</strong>) associates with etiology <strong>of</strong> this lymphoma and is detectable in the<br />

tumor cells.This lymphoma is characterized by the remarkable necrosis formation and the extensive<br />

infiltration <strong>of</strong> inflammation cells in tumor tissue histopathologically. In terms <strong>of</strong> these, we investigate the<br />

interactions between inflammation cells and NNKTL in this study.<br />

Methods: Monocyte and granulocyte were separated from buffy coat <strong>of</strong> the volunteers as inflammation<br />

cells. Each <strong>of</strong> these cells was co-cultured with SNK6 (NNKTL cell line) and KAI3 (<strong>EBV</strong> positive NK cell<br />

line) without IL-2. Proliferation and LMP1 expression were compared with these lines (SNK6, KAI3)<br />

without monocyte culture with/without IL-2. Finally, we investigated CD14 expression <strong>of</strong> 20 NNKTL<br />

patients tissues by immunohistochemistry.<br />

Results: CD14 expression was detected all NNKTL patient tissue. SNK6 and KAI3 showed the<br />

proliferation under the condition that they co-cultured with monocyte without IL-2. LMP1 expression <strong>of</strong><br />

these cells increased at the same time. SNK6 and KAI3 did not show any proliferation under the condition<br />

that they co-cultured with granulocyte without IL-2. LMP1 expression <strong>of</strong> these cells did not increase.<br />

SNK6 and KAI3 did not show any proliferation under the condition that they co-cultured with monocyte<br />

without cell-to-cell contact without IL-2. LMP1 expression <strong>of</strong> SNK6 and KAI3 did not increase at the<br />

same time.<br />

Conclusions: This study suggested that monocyte is one <strong>of</strong> the factors that influence to NNKTL<br />

proliferation. It is possible for NNKTL to proliferate without IL-2, and cell-to-cell contact <strong>of</strong> monocyte<br />

and NNKTL cell was needed to give rise to this phenomenon.<br />
