Linearitet<br />
<strong>Assay</strong>-lineariteten ble bestemt ved å teste fortynnelsesgjenfinningen av en rekke etylglukuronidprøver<br />
i assayet. En urinprøve inneholdende 2000 ng/ml etylglukuronid ble fortynnet i rekkefølge med EtG-fri<br />
urin trinnvis i 25 % inkrementer fra grenseverdikalibratorer. Disse prøvene ble testet i assayet i både<br />
kvalitative og semikvantitative moduser. Alle prøvene ble gjenfunnet innen ±20 % av forventede<br />
verdier i semikvantitativ modus og forventet verdi (mA/min) i kvalitativ modus. Dette indikerer at<br />
assayet er lineært opp til 2000 ng/ml.<br />
Nøyaktighet<br />
184 prøver ble analysert i DRI ® etylglukuronid-assayet både i kvalitative og semikvantitative moduser<br />
og resultatene ble sammenlignet med LC/MS/MS-metoden. I både kvalitativ og semikvantitativ modus,<br />
var enigheten mellom DRI ® EtG-assay og LC/MS/MS 96 % for de positive prøvene. Resultatene<br />
oppnådd av både de kvalitative og semikvantitative modusene oppsummeres nedenfor.<br />
Kvalitativ: Ut av 184 prøver med en grenseverdi på 500 ng/ml, ble 94 prøver detektert som<br />
positive og 85 prøver som negative, og med en grenseverdi på 1000 ng/ml, ble 44 prøver detektert<br />
som positive og 138 prøver detektert som negative av både immunoassayet og LC/MS/MS.<br />
Den totale overensstemmelsen mellom immunoassayet og LC/MS/MS var 97 %. Det fantes fem<br />
uoverensstemmende prøver ved 500 ng/ml-grenseverdien og to uoverensstemmende prøver ved<br />
1000 ng/ml-grenseverdien.<br />
