Projek ini telah bermula sejak September 2009, bertujuan untuk mengkaji status genetik Gajah di Semenanjung<br />

Malaysia dengan menggunakan teknik non-invasive iaitu mendapatkan DNA daripada najis ataupun bulu. Setakat<br />

ini, penganalisaan DNA telah dilakukan terhadap sampel darah Gajah yang diperolehi daripada operasi tangkap<br />

dan pindah dengan memfokuskan kepada dua gen pada DNA mitokondria iaitu Cytochrome B dan Control Region<br />

(D-loop). Sementara itu, optimasi PCR untuk microsattellites markers untuk pembezaan individu dan pembezaan<br />

jantina melalui kaedah molekul sedang dilaksanakan. Melalui analisa DNA dengan menggunakan dua gen pada<br />

mitokondria tersebut, keputusan analisa menunjukkan Gajah-gajah di Semenanjung Malaysia masih terkelompok<br />

dalam kumpulan populasi yang sama dengan perbezaan genetik yang tidak nyata (not significance). Namun begitu,<br />

penganalisaan dengan menggunakan gen-gen lain perlu dilakukan untuk mendapatkan keputusan yang lebih<br />

kukuh. Analisa DNA dengan menggunakan microsattellites markers diharapkan akan memberikan keputusan yang<br />

lebih kukuh dan dapat membezakan antara individu-individu gajah untuk tujuan penentuan saiz populasi Gajah di<br />

Semenanjung Malaysia.<br />

The project was embarked since September 2009, aims to evaluate the genetic status <strong>of</strong> the Elephant population<br />

in Peninsular Malaysia through non-invasive technique to acquire DNA samples from dung <strong>and</strong> hair. To date, DNA<br />

analysis has been done using the blood samples taken from the translocation programme which was focusing on 2<br />

regions <strong>of</strong> mitochondrial genes, the Cytochrome B <strong>and</strong> the Control Region (D-loop). On the other h<strong>and</strong>s, optimizations<br />

<strong>of</strong> PCR on microsatellites markers to differentiate individual <strong>and</strong> molecular sexing markers are currently on progress.<br />

The result from the analysis using the mitochondrial DNA genes shows that there is no significance difference in<br />

genetics among the Elephant population in Peninsular Malaysia. Nevertheless, more studies have to be done using<br />

different genes to give more substantial result. DNA analysis using the microsatellites markers hopefully can give<br />

ample result <strong>and</strong> will be able to differentiate between individual Elephant to assists in the estimation <strong>of</strong> the Elephant<br />

population in Peninsular Malaysia.<br />
