28 �4 Blood sampling an important parameter for prevention of the metabolic syndrome. Gastric inhibitory polypeptide – a specific role in the glycemic index? The glycemic index (GI) classifies carbohydrates with regard to how rapidly the blood glucose level increases after meals. Together with European partners we are currently performing a large prospective study addressing the role of the GI and of protein content on weight regain following weight loss (Diogenes). In this study involving obese families, the subjects first have to reduce body weight by approximately 10 kg using a very low-calorie diet. Subsequently, their diets have a high or low GI or a high or low protein content in four different designs to study the effects on weight and other parameters. The GI is closely associated with the stimulation of incretin hormones such as gastric inhibitory polypeptide (GIP). We observed in humans that GIP is released more rapidly by food with a high GI. GIP unexpectedly modified insulin elimination in the liver, probably by increasing liver perfusion. GIP receptor mRNA expression in adipose tissue of women was inversely associated with insulin levels. GIP also reduced ghrelin release in the postprandial situation. Metabolomics – Complex metabolic patterns as indicators of health Hundreds of metabolites, e.g., carbohydrates, fats, and amino acids, are released by cellular metabolism in the fasting and the postprandial states. Their patterns can now be analyzed by mass spectrometry. We thus expect to identify with high precision the metabolic function and health. We therefore embarked on a characterization of metabolic patterns in cooperation with the Max Planck Institute of Molecular Plant Physiology in Potsdam (Prof. Willmitzer) and the ”Metanomics” Company in Berlin. We have established analytical procedures and are analyzing the MESY BEPO cohort to characterize metabolic patterns associated with obesity, diabetes, and atherosclerosis. �4 Blutentnahme bei einer Studienteilnehmerin Metabolomics – Komplexe Stoffwechselmuster als Indikatoren der Gesundheit Thomas Bobbert, Isabell Göhring, Joachim Spranger Hunderte von Stoffwechselprodukten wie Aminosäuren, Fette, Kohlenhydrate und Zellstoffwechselprodukte werden nüchtern und postprandial von den Organen abgegeben und spiegeln den Zustand des Stoffwechsels wider. Diese können mit modernen Methoden der Massenspektrometrie präzise identifiziert werden. Wir erwarten, dass dadurch in viel genauerem Maße als bisher ein Bild des gesunden individuellen Stoffwechsels gewonnen werden kann. In Kooperation mit dem Max Planck Institut für molekulare Pflanzenphysiologie (Prof. Willmitzer) und der Firma Metanomics in Berlin etablierten wir deshalb diese Technik zur Untersuchung menschlichen Serums und setzten sie in der MESY BEPO-Studie ein. Tatsächlich lassen sich komplexe metabolische Muster identifizieren, die Vorher- sagen über das Diabetes- und Atheroskleroserisiko erlauben. Obwohl aufgrund der großen Zahl von Stoffwechselprodukten komplizierte mathematische Analysen erforderlich sind, ist die Technik viel versprechend und zur Untersuchung der Wirkung von Pflanzeninhaltsstoffen auf den menschlichen Stoffwechsel geeignet.
