2.2 MATERIALS AND METHODS<br />
For generat<strong>in</strong>g appropriate <strong>microbial</strong> consortia towards the envisaged<br />
<strong>ret</strong> <strong>liquor</strong> <strong>treatment</strong> system three types <strong>of</strong> enrichment procedures were<br />
executed as follows:<br />
2.2.1. Successive enrichment <strong>in</strong> husk <strong>in</strong>fusion<br />
2.2.1a Preparation <strong>of</strong> husk <strong>in</strong>fusion<br />
Husk <strong>in</strong>fusion was prepared by boil<strong>in</strong>g 500g freshly split husk <strong>in</strong><br />
20ppt seawater for 30 m<strong>in</strong>utes. The supematant was decanted <strong>of</strong>f and , the<br />
sal<strong>in</strong>ity determ<strong>in</strong>ed and corrected to 20ppt by add<strong>in</strong>g distilled water. For<br />
prepar<strong>in</strong>g the enrichment medium 225ml husk <strong>in</strong>fusion was supplemented<br />
with NHtCl (2gm) , NH 4 NO] (0.5gms) and yeast autolysate (250mg) and<br />
altogether autoclaved at 15 lbs for l Sm<strong>in</strong>, The system was completed by<br />
<strong>in</strong>oculat<strong>in</strong>g the medium with 25ml sample prepared out <strong>of</strong> sediment obta<strong>in</strong>ed<br />
from a <strong>ret</strong>t<strong>in</strong>g ground at Chellanum north. The sediment sample (20gm)<br />
from the above <strong>ret</strong>t<strong>in</strong>g ground was suspended <strong>in</strong> 20ppt sterile seawater<br />
(IOOml) and agitated vigorously for 5 m<strong>in</strong>utes. After allow<strong>in</strong>g the sediment<br />
to settle for a brief period 25ml supematant was <strong>in</strong>oculated <strong>in</strong> 225ml husk<br />
<strong>in</strong>fusion medium. The preparation after adjust<strong>in</strong>g the pH to 7 was<br />
