2.2.7.b CelIulolysis<br />

Cellulolytic potential <strong>of</strong>the segregated cultures were tested <strong>in</strong> m<strong>in</strong>eral<br />

base medium supplemented with filter papers. The m<strong>in</strong>eral base medium<br />

hav<strong>in</strong>g the composition as detailed under section 2.2.3. was dispensed <strong>in</strong> test<br />

tubes at 10ml aliquots and weighed strips <strong>of</strong> filter papers, one set <strong>of</strong><br />

Whatman No.1 and another set <strong>of</strong> ord<strong>in</strong>ary filter paper, were <strong>in</strong>troduced <strong>in</strong><br />

such a way that 0.5cm <strong>of</strong> filter paper was project<strong>in</strong>g out <strong>of</strong> the medium. The<br />

tubes were autoclaved at 10 Ibs for IOm<strong>in</strong> and <strong>in</strong>oculated from a nutrient<br />

agar grown young culture (24 hour growth). The <strong>in</strong>oculated tubes were<br />

<strong>in</strong>cubated at 28±OAoC for 10 days. Whenever evaporation and subsequent<br />

lower<strong>in</strong>g <strong>of</strong> the level <strong>of</strong> medium was observed the loss <strong>of</strong> water was<br />

compensated by add<strong>in</strong>g distilled water. Un<strong>in</strong>oculated controls were also run<br />

along with the test.<br />

On completion <strong>of</strong> 10 days <strong>in</strong>cubation the tubes were observed for<br />

visible degradation <strong>of</strong> the filter paper, loss <strong>of</strong> weight and reduc<strong>in</strong>g sugar<br />
