2.2.7.e. Lipolysis<br />

Tween compounds are generally used as the substrate for detect<strong>in</strong>g the<br />

hydrolysis <strong>of</strong> fats. In addition to tween, the medium must conta<strong>in</strong> a soluble<br />

calcium salt so that the released fatty acid is then turned to <strong>in</strong>soluble calcium<br />

salt around the colony. The medium conta<strong>in</strong>ed the follow<strong>in</strong>g <strong>in</strong>gredients.<br />

The medium was autoc1aved and poured <strong>in</strong>to plates and the organisms<br />

are spot <strong>in</strong>oculated and <strong>in</strong>cubated at 28±OAoC for 7days. Opaque zone<br />

surround<strong>in</strong>g <strong>microbial</strong> growth consisted <strong>of</strong> calcium salts <strong>of</strong> free fatty acid<br />

and are usually taken as be<strong>in</strong>g <strong>in</strong>dicative <strong>of</strong>a positive lipolytic activity.<br />

2.2.7.£ . Amylolysis<br />

Amylase production was checked <strong>in</strong> nutrient agar supplemented with<br />

2% starch. The medium was autoc1aved and poured <strong>in</strong>to plates and spot<br />

<strong>in</strong>oculated and <strong>in</strong>cubated for 72 hours.<br />
