d. Determ<strong>in</strong>ation<br />

20. The optimized conditions <strong>for</strong> the analysis used <strong>in</strong> the CPHL Laboratory are:<br />

• Mobile phase: Methanol:water (89:11)<br />

• Flow rate: 1.3 mL/m<strong>in</strong><br />

• Wavelength: 323 nm<br />

• Retention time: Around 11-12 m<strong>in</strong>.<br />

21. Start HPLC system and allow to warm up and equilibrate <strong>for</strong> m<strong>in</strong>imum <strong>of</strong> 30 m<strong>in</strong> with mobile phase<br />

flow<strong>in</strong>g. Flow rate should be 1.3 mL/m<strong>in</strong>.<br />

22. Inject vitam<strong>in</strong> A standard onto vitam<strong>in</strong> A HPLC system. Adjust mobile phase to achieve a resolution<br />

<strong>of</strong> 1.5 or better <strong>for</strong> cis and trans <strong>for</strong>ms. All trans ret<strong>in</strong>ol should elute <strong>in</strong> approximately 6 m<strong>in</strong> or longer.<br />

23. Inject the standards. Repeat <strong>in</strong>jection <strong>of</strong> standards until peak height(s) or areas are reproducible.<br />

24. Inject sample solutions. Intersperse with standard solution <strong>in</strong>jections after every n<strong>in</strong>e samples to assure<br />

accurate quantitation. (If ret<strong>in</strong>ol peak height or area exceeds the one <strong>for</strong> the high standard by more than<br />

25%, dilute sample solutions us<strong>in</strong>g a solution <strong>of</strong> 10 mL 50% KOH solution, 40 mL 95% ethanol, 10<br />

mL glacial acetic acid, and 40 mL 50:50 THF: ethanol solution).<br />
