VIII. Procedure<br />

A. Stra<strong>in</strong> propagation<br />

1. In aseptic conditions, take a m<strong>in</strong>imum quantity <strong>of</strong> the freeze-dried stra<strong>in</strong> and <strong>in</strong>oculate <strong>in</strong> Trypticase<br />

soy broth. Incubate 24 hours at 37 o C.Enterococcushirae is a facultative anaerobic bacteria. The<br />

growth will look as a precipitate at the bottom <strong>of</strong> the tube (see picture below).<br />

2. After the bacteria have grown <strong>in</strong> agar <strong>for</strong> ma<strong>in</strong>tenance (Part VIII.B), discard the vial.<br />

B. Stra<strong>in</strong> ma<strong>in</strong>tenance (Stock culture)<br />

Once the microorganism has been reconstituted and is grow<strong>in</strong>g adequately, prepare fresh stab culture<br />

every week. Bacteria grow<strong>in</strong>g <strong>in</strong> this culture are the source <strong>of</strong> <strong>in</strong>oculum <strong>for</strong> the analysis. Do not use <strong>for</strong><br />

prepar<strong>in</strong>g <strong>in</strong>oculum if it more than one week old.<br />

1. Take a portion <strong>of</strong> the reconstituted stra<strong>in</strong> and cultivate it <strong>in</strong> a tube with agar <strong>for</strong> stra<strong>in</strong> ma<strong>in</strong>tenance<br />

used <strong>in</strong> the microbiological assay <strong>for</strong> vitam<strong>in</strong>s (HiMedia 132D) and <strong>in</strong>cubate 24 h at 37°C. The<br />

bacteria will look as shown <strong>in</strong> the picture below.<br />

2. Store at 4-8°C. Repeat this procedure weekly. Be<strong>for</strong>e us<strong>in</strong>g new culture <strong>in</strong> assay, make several<br />

successive transfers <strong>of</strong> culture <strong>in</strong> 1-2 week period. From the ma<strong>in</strong>tenance tube, prepare the <strong>in</strong>oculum<br />

<strong>for</strong> the analysis <strong>of</strong> folic acid <strong>in</strong> food.<br />

C. Preserv<strong>in</strong>g the stra<strong>in</strong> <strong>for</strong> a long time<br />

It is recommended to have a stock culture, to get the bacteria at any time. The stra<strong>in</strong> is preserved <strong>in</strong><br />

the stock culture <strong>for</strong> over three months.<br />
