8/21/2012 Lessons learned When planning the use of tissue specimens in research, think like a pathologist and consider how gross, histologic, and quality factors in the tissue may impact your studies “Trust but verify” – be suspicious of any diagnosis or interpretation attached to an outside tissue, unless you have examined it histologically Pathologists and tissue microarrays (TMAs) • Tissue arrays provide rapid screening for dozens or hundreds of tissue specimens with probes for protein, RNA or DNA targets, without exhausting tissue resources. • Traditional TMA = paraffin block • TMAs adaptable to any assay routinely done on tissue sections • Multiorgan TMAs and matching tumor/normal TMAs enhance the output of information 28

Cores of tissue are transferred from donor blocks to the arrayed recipient block. The arrayed slides can be stained by IHC, ISH or FISH. 29
