Kunming Institute of Botany, Chinese Academy of SciencesAnnual ReportN-3-oxo-decanoyl-L-homoserine-lactone Activates Auxin-inducedAdventitious Root Formation via H 2 O 2 - and Nitricoxide-dependent Cyclic GMP Signalling in Mung BeanN-acyl homoserine-lactones (AHLs) are bacterialquorum-sensing signaling molecules that regulatepopulation density. Recent evidences demonstrate theirroles in plant defense responses and root development.H 2 O 2 , nitric oxide (NO) and cyclic GMP (cGMP) areessential messengers that participate in various plantphysiological processes, but how these messengersmodulate the plant response to AHL signals remainspoorly understood.Researchers at KIB show that the N-3-oxodecanoyl-homoserine-lactone(3-O-C10-HL), in contrastto its analogue with an unsubstituted branch chain atthe C3 position, efficiently stimulated the formation ofadventitious roots and the expression of auxin-responsegenes in explants of mung bean seedlings. This responsewas mimicked by the exogenous application of auxin,H 2 O 2 , NO or cGMP homologues, but suppressed bytreatment with scavengers or inhibitors of H 2 O 2 , NOor cGMP metabolism. The 3-O-C10-HL treatmentenhanced auxin basipetal transport; this effect couldbe reversed by treatment with H 2 O 2 or NO scavengersbut not by inhibitors of cGMP synthesis. Inhibiting3-O-C10-HL-induced H 2 O 2 or NO accumulationimpaired auxin- or 3-O-C10-HL-induced cGMPsynthesis, however, blocking cGMP synthesis did notaffect auxin- or 3-O-C10-HL-induced H 2 O 2 or NOgeneration. Additionally, cGMP partially rescued theinhibitory effect of H 2 O 2 or NO scavengers on 3-O-C10-HL-induced adventitious root development and auxinresponsegene expression. These results suggest that3-O-C10-HL, unlike its analogue with unmodifiedbranch chain at the C3 position; can accelerate auxindependentadventitious root formation possibly viaH 2 O 2 - and NO- dependent cGMP signaling in mungbean seedlings. The study has been supported by the Hundredsof Talents Program of CAS and the National NaturalScience Foundation of China, and has been published inPlant Physiology (2012, 158:725-736).The effects of different inhibitors or scavengersand donors on 3-O-C10-HL-induced accumulationof H 2 O 2 and NO in mung bean explants. —— HIGHLIGHTS OF ACHIEVEMENTS ——
