4.3.2 Maturation timing <strong>of</strong> cAMP treated oocytes<br />

for TI<br />

enucleation<br />

Although CHX gave increased cell numbers, cAMP was selected to synchronise<br />

oocytes based on greater expansion <strong>of</strong> cumulus cells which could be beneficial for<br />

cytoplasmic maturation, higher efficiency <strong>of</strong> synchronising porcine IVM and higher<br />

percentage <strong>of</strong> MII oocytes at 44 hpm and the frequency <strong>of</strong> parthenogenetic<br />

development which did not differ from CHX treated or control groups. <strong>The</strong><br />

distribution <strong>of</strong> nuclear morphology <strong>of</strong> oocytes cultured in the absence or presence <strong>of</strong> 1<br />

mM cAMP was examined at 2h intervals between 38 hpm and 44 hpm. <strong>The</strong> entire<br />

meiotic progression <strong>of</strong> oocytes in control and cAMP group was shown in Figure 4.2,<br />

respectively. Pictures were derived based on the data from Table 4.2 combined with<br />

data from section 3.3.3. It was found that TI oocytes peaked at 38 hpm after cAMP<br />

treatment.<br />
