the percent <strong>of</strong> polymorphism present, which equals the nurnber <strong>of</strong> bands that were<br />

polymorphic divided by the <strong>to</strong>tal number <strong>of</strong> bands. Primer 318 was not cansidered<br />

because it produced only monornorphic bands in the DSC-3 DH Iines.<br />

<strong>The</strong> number <strong>of</strong> individuals analyzed in C, and C, was deteminecl by the<br />

frequency <strong>of</strong> bands expressd in the OH lines (Table 5.3). <strong>The</strong> lowest band frequency<br />

(p) with a specific primer was calailated by dïviding the number <strong>of</strong> indhiduals with the<br />

least frequently expressed band by the <strong>to</strong>tal number <strong>of</strong> individual Iines in the DH<br />

population. <strong>The</strong>n the number <strong>of</strong> plants (N) sampled fmm C, and C, was calculated <strong>to</strong><br />

give a 95% probability (P) that the least frequent allele (p) would be represented, as<br />

shown in the following equation (Jasieniuk et al., 1996):<br />

RAPD fragment sizes were estimated f<strong>to</strong>m the gels in relation <strong>to</strong> the size marker.<br />

Data matrix tables were compiied where bands were recorded as either present (1) or<br />

absent (O), as show in Table 5.4 and Appendix 11-14.<br />

RESULTS AND DISCUSSION<br />

RA PD Analysis<br />

Polyrnorphism was detected for 9 <strong>of</strong> the original 30 primers tested between the<br />

DSC-3 and Reward donor populations (data not shown). When tested on the DH lines<br />

in both populations, 5 prime- produced bands that expressed adequate intensity <strong>to</strong>
