We also characterizedthe GUS staining patterns in transgenic flowers (Table 3). Among the 133 lines thatshowed GUS activity in flowers, 50 lines (37.6%) displayed intense GUS stainingprimarily in the palea and lemma. One line exhibited GUS activity only in glumes,eight lines showed GUS activity only in lodicules, and four lines only in a carpel.Of the 11 lines exhibiting stamen-specific GUS activity, seven showed pollen-specificGUS staining. Five to ten days after flowering, the developing seeds were alsosubjected to GUS staining. A large portion of these lines showed a tissue-preferentialexpression pattern. For example, line G930726 exhibited an aleurone-layer preferentialGUS staining pattern, indicating that the trapped gene might be involved in theformation of the aleurone layer or in a specific function in the tissue.DiscussionA variety of GUS staining patterns was observed from the tagged lines. Some weretissue- or organ-specific and others were expressed ubiquitously. This observationsupports T-DNA insertion as a random event. The flanking sequences of the GUSpositivelines are being isolated to obtain the genes that provided GUS expression.Generation of T-DNA insertional tagging lines in rice 259
