Fast retrieval avoidsprolonged thaw cycles that limit theviability of cells and can also ruin otherbiological samples.CryogenicsThe cryobank vials are arrayed ina microwell format that allows forthe use of multi-channel pippetorsor liquid handling robots. Thisfacilitates sterilework and minimizes handling time.The racks are selected in the databaseprogram. Each rack can contain more vialsthan are required to be picked.The Nunc tube selection tool is intended forpicking several vials out of a rack. While stillat room temperature, the bits are placed inthe positions listed in the database selectionlist. After verifying the bit position, therack is ready to be taken out of the cold.Caps are replaced using the singleor multiple head screwdrivers.Vials are locked into the rack,which ensures that they do not fallout during transport in the lab orstorage. The snap in the rack alsoretains the vial in the rack when thecap is mounted and the screwdriveris removed.The cryobank rack position is listed in thedatabase output and can be verifed byscanning the barcode on the cryobankrack.We guarantee verified uniquebarcodes on the racks and on thecryobank vials. 2D codes are laseretched into the bottom of the vial.The 2D codes are robust and offerthe ability to read multiple codesat the same time using the Nuncscanner.The scanner software reads thecodes and sends vial coordinateinformation to a databaseprogram.The Nunc tube selection tool has a cut-offcorner that ensures that the rack and tubeselection tool orientation is always alignedwhen the levers are pressed out of the rackand lifted up for easy retrieval. The rest ofthe rack is rapidly returned to the freezer toavoid significant thawing.The database program uses thelocation of the vials in the rack, thelocation of the rack in the freezer,and the associated sample data tokeep track of the position of everysample and combine it with thedata used for selecting samplesat retrieval.Codes can be verified either prior to or afterthaw. If the database is used properly, theuser avoids problems with condensation orfrosting during the reading process, as thetubes can thaw prior to checking the codes.Condensation and frost are problems thatcan be overcome by wiping the codes,having them covered during storage orusing dehumidified air.Nunc racks are available for bothdewars and freezers that optimizethe storage density. Racks aremade in aluminum and can becolor coded. Formats are bothfor microplate format racks andstandard cryobox formats.Vials should always be checked to makesure that actually the correct samples havebeen retrieved.51
