observed and the titer was found to be 1600<br />

and 800AU/ml respectively. Finally, to characterize<br />

the inhibitory agents, tests were conducted<br />

on FII24. Since bacteriocins are by<br />

definition proteinaceous substances they must<br />

be sensitive to at least one proteolytic enzyme.<br />

Consequently, protease sensitivity is a key criterion<br />

in their characterization. In our case, αchymotrypsin,<br />

trypsin or proteinase K had no<br />

effect on activity of FII83, whereas the<br />

inhibitory activity of FII24 was compl<strong>et</strong>ely<br />

lost by the same enzymes, reflecting the proteinaceous<br />

nature of the inhibitory agent produced<br />

by Rhizobium sp. ORN24 strain.<br />

Additionally, compl<strong>et</strong>e inactivation of FII83<br />

was observed when it was exposed to 100oC for 20 min, while FII24 showed resistance to<br />

heat, r<strong>et</strong>aining all activity after exposure to<br />

100 oC for 60 min.<br />

The other physico-biochemical tests showed<br />

that both FII83 and FII24 r<strong>et</strong>ained their antibacterial<br />

activity within the range of pH 4 to<br />

7 and maintained full stability after storage for<br />

60 days at 4 oC, 0 oC and at <strong>–</strong> 20 oC, while no<br />

activity was d<strong>et</strong>ected after storage for 80 to<br />

120 days at 37 oC. Tween 80, Tween 20, SDS<br />

or Triton X-100 seems to have a marked effect<br />

on FII24; in contrast the activity of FII83 was<br />

not reduced when treated with these agents.<br />

As <strong>des</strong>cribed in Materials and m<strong>et</strong>hods, samples<br />

were taken from inhibition zones and surface<br />

staked to medium agar plates. After incubation<br />

of plates, growth of bacteria was not<br />

observed. This result suggests that the antibacterial<br />

agents of both FII83 and FII24 are<br />

bactericidal toward the targ<strong>et</strong> strain. Additionally,<br />

strains were resistant to their own cell<br />

free supernatants. This may be due to an<br />

immunity substance simultaneously being<br />

produced by the producer strain along with its<br />

product, as in the case of some bacteriocins<br />

(Kacem 2007).<br />

As shown in Table 2, the inhibitory action of<br />

FII24 was tested against a total of 37 bacteria<br />

represented by 6 strains of Rhisobium,<br />

8 strains of Sinorhizobium, 10 strains of<br />

E. chrysanthemi, 7 strains of L. plantarum,<br />

4 strains of P. freudenriechii shermanii,<br />

3 strains of Pseudomonas, and one strain of<br />

E. coli FII24 showed inhibition against Rhizobium<br />

sp. (ORN19 and ORN23) and<br />

Sinorhizobium sp. (ORN17, ORN20, and<br />

ORN22) among the 14 strains tested.<br />

Inhibitory activity was also directed against<br />

the natural flora present in soil, including<br />

Pseudomonas (Pseudomonas syringae).<br />

P. syringae is a group comprising more than<br />

50 plant pathogens classified as pathovars on<br />

the basis of host specificity (Young <strong>et</strong> al.<br />

1996).<br />

Discussion<br />

In the present investigation we report the production<br />

and characterization of an antimicrobial<br />

substances present in the cell free supernatants;<br />

FII83 and FII24 respectively,<br />

obtained from Rhizobium sp. ORN83 and<br />

ORN24 strains isolated in saline soils in the<br />

region of Oran located in western Algeria.<br />

Among the six strains screened for their<br />

antagonistic activity, FII83 and FII24 from<br />

Rhizobium sp. ORN24 and ORN83 strains<br />

showed respectively, an inhibition zone of<br />

10 mm and 25 mm in diam<strong>et</strong>er on indicator<br />

strain. Similar results have been reported by<br />

Hirsch (1979) and Rodelas <strong>et</strong> al. (1998).<br />
