Chapter 4AbstractMeiotic abnormalities were <strong>in</strong>vestigated <strong>in</strong> <strong>in</strong>terspecific lily hybrids us<strong>in</strong>g genomic <strong>in</strong> situhybridization (GISH) and fluorescence <strong>in</strong> situ hybridization (FISH). At metaphase I, thebivalents <strong>in</strong>volv<strong>in</strong>g homoeologous <strong>chromosome</strong>s and unpaired univalent were the ma<strong>in</strong>configuration for most <strong>of</strong> the pollen mother cells. Besides these, also multivalents as well asbivalents <strong>in</strong>volv<strong>in</strong>g non-homologous <strong>chromosome</strong> pair<strong>in</strong>g <strong>in</strong> the same genome, were alsoobserved. Moreover, broken <strong>chromosome</strong>s were sporadically detected at metaphase I us<strong>in</strong>gGISH and FISH with telomere repeats as probe. At anaphase I, chromatid bridgesaccompanied with fragments were present. GISH and FISH revealed that these bridges<strong>in</strong>volved not only non-sister chromatids but also sister-chromatids. This strongly suggests thatthe bridges and the fragments found were derived from U-type exchanges. In conclusion, U-type exchanges, <strong>in</strong>clud<strong>in</strong>g spontaneous chromatid breakage and fusion, leads to anaphasebridg<strong>in</strong>g at meiosis <strong>in</strong> <strong>in</strong>terspecific hybrids <strong>of</strong> lily. It is argued that dur<strong>in</strong>g meiosis <strong>of</strong><strong>in</strong>terspecifc hybrids <strong>of</strong> lily, both homologous recomb<strong>in</strong>ation (HR) and nonhomologous endjo<strong>in</strong><strong>in</strong>g (NHEJ) were both <strong>in</strong>volved to repair double strand breaks (DSBs).
