3.1.3 PAO1-lys10 and PA14-lys10<br />

<strong>The</strong> genomic island at <strong>the</strong> lys10 tRNA locus with<strong>in</strong> P. aerug<strong>in</strong>osa PA14 is <strong>the</strong><br />

pathogenicity island, PAPI-2 (He et al. 2004). <strong>The</strong> P. aerug<strong>in</strong>osa PAO1 has a<br />

genomic island present at this tRNA site locus def<strong>in</strong>ed <strong>in</strong> <strong>the</strong> literature (He et al.<br />

2004), which has some sequence similarity with PAPI-2.<br />

<strong>The</strong> capture vector conta<strong>in</strong><strong>in</strong>g <strong>the</strong> PAPI-2 island (PA14-lys10) was used<br />

subsequently <strong>in</strong> complementary studies performed by Ewan Harrison (Harrison E,<br />

PhD <strong>the</strong>sis, University <strong>of</strong> Leicester, unpublished) <strong>in</strong>volv<strong>in</strong>g <strong>the</strong> P. aerug<strong>in</strong>osa<br />

pathogencity island deletant mutants, PA14 ∆PAPI-2 and PA14 ∆PAPI-1∆PAPI-2.<br />

Figure 3-3 shows <strong>the</strong> restriction pr<strong>of</strong>ile <strong>of</strong> both PAO1-lys10 and PA14-lys10 us<strong>in</strong>g<br />

I-SceI.<br />

Figure 3-3 shows a 0.6% agarose gel with <strong>the</strong> restriction pr<strong>of</strong>ile <strong>of</strong> PAO1-lys10 and PA14-lys10<br />

<strong>The</strong> genomic island present with<strong>in</strong> P. aerug<strong>in</strong>osa PA14 at <strong>the</strong> lys47 tRNA site is<br />

PAPI-1, <strong>the</strong> o<strong>the</strong>r pathogenicity island explored dur<strong>in</strong>g <strong>the</strong> mur<strong>in</strong>e studies with<strong>in</strong><br />

this <strong>the</strong>sis. <strong>The</strong> successful captur<strong>in</strong>g <strong>of</strong> this island would have been useful for<br />

complementary studies with <strong>the</strong> pathogenicity island deletant mutants.<br />

Unfortunately this island was never successfully captured. A set <strong>of</strong> experiments<br />
