SPM5 and the DARTEL method were used to perform a VBM analysis to assess WM differences in 14 early onset AD (EOAD) and<br />

15 late onset AD (LOAD) vs. age- and gender-matched healthy controls (HC). Compared to HC, in EOAD patients, WM loss was<br />

mapped mainly to the posterior regions such as the posterior cingulum and the lateral parietal regions, bilaterally. In LOAD patients,<br />

WM loss was confined to the medial temporal lobe in the parahippocampal regions. Our findings indicate that EOAD and LOAD<br />

patients differ in the topography of WM damage, which reflects the pattern of cortical loss.<br />

The diffusion profile in semantic dementia (SD) was studied to examine the profile of connection changes. Tract-based spatial<br />

statistics in SD patients provided compelling evidence that the network is distinct to that of Alzheimer’s disease (AD), and indicated<br />

that the key abnormality exiting the temporal lobe was in the uncinate fasciculus projection to the orbital frontal lobe. The tensor<br />

behaviours also indicated that the nature of the neurodegenerative in SD differs qualitatively from that seen in AD.<br />

Measurement of lateral ventricular volume is often used for measuring the progression of neurodegenerative diseases. Existing<br />

techniques for ventricle segmentation have been extensively validated for the normal population, but may be suboptimal for the<br />

patients with Alzheimer’s Disease (AD), for example. We propose an automated method which uses information from expert manual<br />

segmentation combined with a population-specific atlas. Experiments were completed with a group of 271 elderly patients from an<br />

ongoing clinical trial, using manual segmentations as a gold standard. We found that proposed algorithm yields accurate results with a<br />

median kappa of 0.962.
