Introduction générale<br />
Les dérivés benzénique 1,2ether énolique polyfonctionnalisée pressentent<br />
un intérêt considérable en synthèse organique du fait <strong>de</strong> leur présence dans<br />
certains molécules, dont les applications sont nombreuses et variées (<br />
pharmacologique ,biologique , cosmétique, agriculture,…… .).<br />
Dans ce contexte, nous nous sommes intéressés à la préparation <strong>de</strong> dérivés<br />
<strong>de</strong> structure similaire. en partant <strong>de</strong> trois composés <strong>de</strong> bon marché et<br />
disponibles, le β-cétoesters le β-dicétones , et l'aldéhy<strong>de</strong> salicylique.<br />
Par ailleurs, l'accès à ses composés fait intervenir <strong>de</strong>s réactions simples<br />
d'alkylation qui se déroulent dans <strong>de</strong>s conditions douces.<br />
La stratégie retenue pour accé<strong>de</strong>r à ces composés comporte trois gran<strong>de</strong>s<br />
étapes :<br />
- Alkylation <strong>de</strong>s β-cétoesters (β-dicétones ) par les enchaînement insaturés<br />
allylique et propargylique .<br />
- O-alkylation <strong>de</strong> l'aldéhy<strong>de</strong> salicylique par les agent alkylants ( bromure<br />
d'allyle et propargyle, la 4-bromo but 1-éne , le 5-bromopent-1-éne ).<br />
- Con<strong>de</strong>nsation <strong>de</strong>s β-cétoesters et <strong>de</strong>s β-dicétones alkylés sur les aldéhy<strong>de</strong>s<br />
salicylique alkylés.

Nos travaux <strong>de</strong> recherche sont présentés dans ce mémoire suivant un plan divisé<br />
en cinq grands chapitres .
Chapitre1 : le premier chapitre sera consacré à donner quelque rappels<br />
bibliographiques sur les différentes métho<strong>de</strong>s <strong>de</strong> synthèse <strong>de</strong>s squelettes benzénique<br />
1,2 éther énolique polyfonctionnalisée nous décrivons aussi dans ce chapitre la<br />
réactivités <strong>de</strong> leurs squelettes <strong>de</strong> base ( phénol, benzal<strong>de</strong>hy<strong>de</strong>, dérivés β-dicarbonylés<br />
).<br />
Chapitre 2 : synthèse <strong>de</strong> β-cétoesters alkylés et β-dicétones alkylés .<br />
Chapitre 3 : O-alkylation <strong>de</strong> l'al<strong>de</strong>hy<strong>de</strong> salicylique .<br />
Chapitre 4 : synthèse <strong>de</strong> composés benzénique1, 2 éther énolique<br />
polyfonctionnalisée.<br />
Une conclusion générale sera donneé à la fin <strong>de</strong> ce mémoire rassemblent les les<br />
principaux résultas obtenus au cours <strong>de</strong> notre travail.<br />
Les différents mo<strong>de</strong>s operatoires utilisés ainsi que les principales caractéristiques<br />
spectroscopiques ( IR , RMN 1 H, RMN 13 C ) <strong>de</strong>s composés obtenus seront regroupés<br />
dans la partie expérimentale du 5 éme chapitre.

Les dérives benzéniques 1,2-polyfonctionnalisés représentent <strong>de</strong>s structures<br />

Des composés dérivant <strong>de</strong> l’eugénol sont utilisés comme pansement <strong>de</strong>ntaire pour leurs<br />

Le choix <strong>de</strong> ce type <strong>de</strong> composés est basé sur :<br />
1- L’intérêt potentiel <strong>de</strong> ces structures dans <strong>de</strong>s domaines très divers dont nous avons donné<br />
un petit aperçu précé<strong>de</strong>mment .<br />
2- La présence au sein <strong>de</strong> ces dérivés <strong>de</strong> plusieurs sites réactionnels (nucléophiles et<br />
électrophiles), pouvant faire l’objet <strong>de</strong> transformations et autres développements très intéressants<br />
(oxydation, réduction, addition, cyclisation, con<strong>de</strong>nsation, etc… ..).<br />

Conclusion :<br />
Dans ce chapitre, nous avons présenté dans un premier temps quelques exemples <strong>de</strong> la<br />
bibliographie <strong>de</strong>s composés <strong>de</strong> structure benzénique 1,2-polyfonctionnalisé avec quelques unes <strong>de</strong><br />
leurs applications dans différents domaines, cosmétique, agriculture, pharmaceutique, etc…<br />
Nous avons ensuite exposé <strong>de</strong>ux métho<strong>de</strong>s <strong>de</strong> synthèse <strong>de</strong> ce type <strong>de</strong> composé, que nous<br />
avons relevé parmi le peu <strong>de</strong> résultats trouvé dans la littérature.<br />
L’analyse rétrosynthétique <strong>de</strong> notre molécule-cible nous a conduit à <strong>de</strong>s intermédiaires<br />
réactionnelles facilement accessibles : dérivés β-dicarbonylés α-alkylés et l’aldéhy<strong>de</strong> o-salicylique O-<br />
alkylé.<br />
Le schéma <strong>de</strong> synthèse que nous retenu ne fait intervenir que <strong>de</strong>s réactions d’alkylation et <strong>de</strong>s<br />
con<strong>de</strong>nsations.
Introduction :<br />
Ce chapitre est consacré à la préparation <strong>de</strong> composés nécessaires pour effectuer l’étape<br />
<strong>de</strong> con<strong>de</strong>nsation.<br />
D’après notre schéma <strong>de</strong> synthèse, ces composés sont <strong>de</strong>s β-céto-esters ou <strong>de</strong>s β-<br />
dicétones α-alkylés et l’aldéhy<strong>de</strong> ortho-salicylique O-alkylé.<br />
Avant d’abor<strong>de</strong>r notre travail <strong>de</strong> synthèse proprement dit, nous avons fait quelques<br />
rappels sur les propriétés chimiques très diverses <strong>de</strong> ces dérivés β-dicarbonylés et l’aldéhy<strong>de</strong><br />
ortho-salicylique, afin <strong>de</strong> montrer leur importance en synthèse organique.<br />

II-2-1-1 Alkylation par le bromure d’allyle :<br />
L’acétyle acétate <strong>de</strong> méthyle 1a réagit avec <strong>de</strong> bromure d’allyle en présence d’un excès<br />
(4eq ) <strong>de</strong> carbonate <strong>de</strong> potassium dans l’acétone. Le mélange réactionnel a été laissé 20 heures<br />
au reflux en suivant l’avancement <strong>de</strong> la réaction par chromatographie sur couche mince (CCM ). Le<br />
2-acétyle pent-4-ènoate <strong>de</strong> méthyle 2a est ainsi synthétisé avec un ren<strong>de</strong>ment <strong>de</strong> 63% après<br />
distillation sous pression réduite .

propargyle dans les mêmes conditions opératoires utilisées pour le bromure d’allyle.<br />

Nous avons isolé, sans difficulté, 65% <strong>de</strong> produit le 2-acétyl-pent – 4-ynoate <strong>de</strong> méthyle 3a<br />

Ces résultats nous permettent <strong>de</strong> faire les quelques remarques suivantes :<br />
Tous les essais <strong>de</strong> l’alkylation <strong>de</strong> β-cétoesters et <strong>de</strong> β-dicétones ont donné <strong>de</strong>s<br />
ren<strong>de</strong>ments,- non optimisés-, intéressants (compris entre 63% et 73%) compte-tenu <strong>de</strong><br />
l’utilisation <strong>de</strong> réactifs commerciaux peux coûteux .<br />
L’analyse spectroscopique IR, RMN 1 H, RMN 13 C, nous a donné <strong>de</strong>s résultats satisfaisants.<br />
Exemple : l’étu<strong>de</strong> spectrale <strong>de</strong> composé 2a<br />
- Le spectre <strong>de</strong> RMN 1 H enregistré à 300MHz du composé 2a représenté ci-après, fait<br />
apparaître <strong>de</strong>s différences très nettes par rapport aux composés <strong>de</strong> départ 1a .<br />
Le spectre montre bien l’apparition<br />

Ces résultas nous ont permis <strong>de</strong> noter quelques observations sur cette étu<strong>de</strong> :<br />
* Dans toutes les expériences que nous avons menées, nous avons isolé les composés<br />
O-alkylés facilement à séparer sur colonne ( chromatographie sur colonne ).<br />
* Les conditions opératoires, K2CO3 dans le méthanol au reflux permet d’accé<strong>de</strong>r et avec un<br />
bon ren<strong>de</strong>ment à <strong>de</strong>s produits obtenus par O-alkylation .<br />
II-2-4-6 Détermination <strong>de</strong> la structure <strong>de</strong>s produits :<br />
Les métho<strong>de</strong>s spectrales classiques, RMN 1 H, 13 C, IR, ont été utilisées pour i<strong>de</strong>ntifier les<br />
composés O-alkylés.<br />
Exemple : le 2-( but-3-ényloxy ) benzaldéhy<strong>de</strong> .<br />
L’analyse par spectroscopie IR à donner les ban<strong>de</strong>s d’absorption suivantes :<br />
3077,85cm -1 élongation <strong>de</strong>s H du phényle.<br />
1707,18cm -1 élongation du C=O <strong>de</strong> l’aldéhy<strong>de</strong> conjugué.<br />
1601,60cm- élongation <strong>de</strong> la C=C <strong>de</strong> C=C non conjugué.<br />
1067,89cm -1 vibration d’élongation du C-O.<br />
997,02cm -1 vibration <strong>de</strong> déformation du plan <strong>de</strong>s liaisons –CH=CH2.<br />
753,55cm -1 vibration hors du plan <strong>de</strong>s liaisons C-H pour <strong>de</strong>s noyaux benzéniques<br />
disubstitués en position ortho.<br />
Dans tous les cas que nous avons étudier nous observons la disparition <strong>de</strong> bon<strong>de</strong> large entre<br />
3590 et 3650 cm -1 revient au vibration d’élongation du OH (alcools) Comparé au spectre du produit<br />
<strong>de</strong> départ.<br />
L’analyse par spectroscopie RMN 1 H permet <strong>de</strong> donner les résultas suivantes :<br />
Le spectre <strong>de</strong> 2- ( but-3-enyloxy) benzaldéhy<strong>de</strong> comparé au spectre <strong>de</strong> produit <strong>de</strong> départ<br />
montre la présence <strong>de</strong> nouveaux signaux . correspondants aux protons allyliques et homoallyliques<br />
(voir <strong>de</strong>scription spectroscopique <strong>de</strong>s composés dans le chapitre 5 partie expérimentale).
Les protons allyliques (-CH=CH2), résonnant entre 6.47 – 6,57 ppm apparaissent sous<br />
forme <strong>de</strong> multiplet.<br />
Les protons allyliques ( -CH2-CH=CH2 ), apparaissent sous forme d’un multiplet et<br />
leurs déplacements chimiques se situent entre 4,68 et 4,74 ppm<br />
Les protons homoallyliques ( -CH2-CH2-CH=CH2 ), apparaissent sous forme d’un<br />
doublet <strong>de</strong> doublet, leurs déplacements chimique se situent à 4,17ppm.<br />
Les protons allyliques géminés.<br />

Le proton Hc résonnant à 6,05 ppm sous forme <strong>de</strong> doublet et le proton Hc’ résonnant à<br />

Les proton He, Hg, Hf , Hh , résonnant entre 7,4 et 8,52 ppm sous forme <strong>de</strong>s multiplets.<br />
L’ analyse par RMN 13 C permet <strong>de</strong> donner les résultats suivantes :<br />
Le spectre <strong>de</strong> composé 2-( but –3 enyloxy ) benzal<strong>de</strong>hy<strong>de</strong> comparé au spectre <strong>de</strong> produit <strong>de</strong><br />
départ montre l’apparition <strong>de</strong>s nouveaux signaux, correspondant aux carbones allyliques et<br />
homoallyliques (voir <strong>de</strong>scription spectroscopiques <strong>de</strong>s produits dans le chapitre 5, partie<br />
Le carbone allylique (-CH=CH2 ) résonne à 140,47.<br />
Le carbone allylique (-CH=CH2 ) résonne à 117,66.<br />
Le carbone allylique ( -CH2-CH=CH2 ) résonne à 69,09.<br />
Le carbone homoallylique ( -CH2-CH2-CH=CH2 ) résonne à 33,98 .<br />

Remarque :
Dans une étu<strong>de</strong> antérieure, similaire à la notre, il a été, également d’isolé avec le substrat<br />
<strong>de</strong> c-alkylation, un produit secondaire issu d’un o-alkylation avec un ren<strong>de</strong>ment compris<br />
entre 5 et 10 % et un autre issu d’une réaction <strong>de</strong> dialkylation avec un ren<strong>de</strong>ment <strong>de</strong> 8% 21 .<br />
L’α-alkylation <strong>de</strong>s β-cétoesters et <strong>de</strong>s β-dicétones par <strong>de</strong>s groupements carbonés insaturés, a<br />
Conclusion :<br />
conduit aux produits alkylés attendus avec <strong>de</strong>s ren<strong>de</strong>ments non optimisés <strong>de</strong> 63 à 73 %. Quant à la<br />
O-alkylation, - également par <strong>de</strong>s groupements carbonés insaturés-, du l’aldéhy<strong>de</strong> ortho-salicylique,<br />
elle a donné <strong>de</strong>s dérivés du l’aldéhy<strong>de</strong> ortho-salicylique O-alkylés avec <strong>de</strong>s ren<strong>de</strong>ments moyens<br />
(mais non optimisés) <strong>de</strong> 50 à 69 %.<br />
Il nous faut signaler que tous nos réactions d’alkylations ont été effectuées dans <strong>de</strong>s<br />
conditions réactionnelles simples et économiques initialement, utilisant le K2CO3 dans l’acétone au<br />
Les produits isolés, après leur purification par chromatographie sur gel <strong>de</strong> silice, ont été tous<br />
reflux.<br />
i<strong>de</strong>ntifiés par les métho<strong>de</strong>s spectroscopiques (IR, RMN- 1 H, - 13 C).<br />
Cette étape d’alkylation nous a permis <strong>de</strong> préparer les réactifs nécessaires à l’étape<br />
principale <strong>de</strong> nos recherches c’est à dire la con<strong>de</strong>nsation <strong>de</strong> ces dérivés.
Introduction :<br />
Ce chapitre regroupe les réactions <strong>de</strong> con<strong>de</strong>nsation <strong>de</strong>s β-cétoesters et β-dicétones α-<br />
alkylés sur les aldéhy<strong>de</strong>s ortho- salicyliques O-alkylés.<br />
La con<strong>de</strong>nsation consiste à faire réagir un β-cétoester ou une β-dicétone α-alkylés avec<br />
un aldéhy<strong>de</strong> ortho-salicylique O-alkylé, dans <strong>de</strong>s conditions réactionnelles faisant intervenir le<br />
DBU dans le MeOH anhydre, à température ambiante durant 15 heures environ.<br />
Le choix <strong>de</strong> ces conditions - DBU dans le MeOH -, est basé sur les résultats <strong>de</strong>s travaux effectués<br />
par Charonnet. 23 Cette étu<strong>de</strong> montre que les β-cétoesters et les β-cétoami<strong>de</strong>s réagissaient avec le<br />
benzaldéhy<strong>de</strong> comme électrophile en présence <strong>de</strong> DBU dans le méthanol, donnant <strong>de</strong> bons résultats<br />
par rapport aux autres expériences effectuées dans <strong>de</strong>s conditions opératoires différentes, K2CO3\<br />
MeOH, MeONa\ MeOH, NaH/THF, DBU\THF, DBU\ toluène, DBU\ MeOH.<br />
Dans ce chapitre, nos résultats sont présentés en <strong>de</strong>ux gran<strong>de</strong>s parties :<br />

Les con<strong>de</strong>nsations, suivies par CCM, du 2-acétyl-pent–4–ynoate <strong>de</strong> méthyle 3a sur le 2-(<br />
allyloxy ) benzal<strong>de</strong>hy<strong>de</strong> 2c , le 2-(but-3-ènyloxy) benzaldéhy<strong>de</strong> 4c et le 2-( pent-4- ènyloxy)<br />
benzaldéhy<strong>de</strong> 5c, nous avons permis d’isoler respectivement les produits con<strong>de</strong>nsés 8a ,9a et 10a<br />
avec <strong>de</strong>s ren<strong>de</strong>ments <strong>de</strong> 54%, 43% et 51%, après chromatographie sur gel <strong>de</strong> silice.<br />

L’examen <strong>de</strong> ce tableau nous montre que :<br />
- Les ren<strong>de</strong>ments non optimisés <strong>de</strong> ces con<strong>de</strong>nsations se situent entre 43 et 64 % .
- Toutes les structures <strong>de</strong>s produits <strong>de</strong> con<strong>de</strong>nsation ont été confirmées par les métho<strong>de</strong>s<br />
d’analyse spectroscopique IR , RMN 1 H et 13 C.<br />
- Ces résultats sont satisfaisants par rapport à <strong>de</strong>s travaux similaires relevés dans la<br />
littérature. 23<br />
En effet, lors <strong>de</strong> la préparation du (E)-5-(2-furyl)-2-méthyl-3-oxopent-4-énoate <strong>de</strong> éthyle c , au<br />
lieu <strong>de</strong> l’obtention du produit c souhaité, les auteurs ont obtenus, - dans les mêmes conditions<br />
opératoires (mais à température plus élevée et une durée plus longue) -, un mélange <strong>de</strong> <strong>de</strong>ux<br />
produits non séparables:<br />

Les auteurs expliquent l’obtention du composé majoritaire d par une séquence γ-aldolisation /<br />
déshydratation, suivie d’une saponification / décarboxylation <strong>de</strong> la fonction ester du substrat <strong>de</strong><br />

A partir <strong>de</strong>s résultats obtenus dans ces con<strong>de</strong>nsations, nous pouvons faire les observations<br />
suivantes :<br />
Les différents essais <strong>de</strong> la con<strong>de</strong>nsation <strong>de</strong>s β-dicétones α-allyliques et α-propargyliques<br />
sur les aldéhy<strong>de</strong>s salicyliques O-alkylés ont donné <strong>de</strong>s résultats satisfaisants <strong>de</strong> point <strong>de</strong> vue<br />
réactivité.<br />
Ces con<strong>de</strong>nsations nous ont permis d’accé<strong>de</strong>r à <strong>de</strong>s composés polyfoncyionnalisés dans <strong>de</strong>s<br />
conditions réactionnelles relativement simples.<br />
Les ren<strong>de</strong>ments observés , - <strong>de</strong> 45 à 71 % -, sont intéressants, puisque non optimisés.<br />
Mécanisme proposé :<br />
D’après la littérature 23 , on peut envisager dans une premiere étape une α - aldolisation<br />
réversible ou un processus d’aldolisation – rétroaldolisation .<br />
Le mécanisme proposé, faisant intervenir , tout d’abord la formation <strong>de</strong> l’α-aldol instable A qui<br />
subirait une rétro-aldolisation pour donner l’énolate B défavorisé cinétiquement et<br />
thermodynamiquement 23,24 . Ce <strong>de</strong>rnier réagit avec l’al<strong>de</strong>hy<strong>de</strong> pour donner un γ-aldol plus stable C<br />
qui après déshydratation, fournit les β-dicétones ( β-cétoesters ) conjugueés α-substituées.<br />

Par ailleurs, il faut signaler que nous n’avons obtenu que <strong>de</strong>s produits correspondant à une seule<br />
orientation <strong>de</strong> la réaction <strong>de</strong> C-alkylation.<br />
Cette régioselectivité peut s’expliquer par la théorie <strong>de</strong> Pearson, faisant intervenir les notions <strong>de</strong><br />
« centre dur – centre mou » <strong>de</strong>s dérivés 1,3–dicarbonylés et <strong>de</strong>s électrophiles. 25<br />
Ce principe dit H.S.A.B. 26 suit une règle simple : « un aci<strong>de</strong> mou préfère se lier avec une base<br />
molle et un aci<strong>de</strong> dur préfère se lier avec une base dure ».<br />
Concernant les sites d’attaque <strong>de</strong> nos produits l’arrachement <strong>de</strong> l’hydrogène avec le DBU qui une<br />
base dure l’attaque se fait sur un hydrogène du carbone terminal plus dur.<br />

Par ailleurs, le caractère « dur-mou » <strong>de</strong>s composés 1,3 – dicarbonylés diffère selon la nature<br />

électronique du substituant en position α <strong>de</strong> la fonction carbonyle .<br />
En effet, les β-dicétones substituées par un groupement alkyle faiblement donneur engendreront<br />
les anions fortement stabilisés qui constituent un centre « mou ».<br />
De même, les β-cétoesters qui portent un groupe alkoxy dont le caractère donneur d’électrons est<br />
un peu plus marqué donnent <strong>de</strong>s anions légèrement moins stabilisés mais qui conservent un<br />

caractère relativement « mou ».<br />
La stabilité <strong>de</strong>s anions correspondants est directement liée à l’acidité relative <strong>de</strong>s protons en α qui<br />
diminue progressivement lorsque nous passons d’une β-dicétone à un β-céto-esters.

Aux termes <strong>de</strong> ce travail , nous avons atteint notre objectif qui consistait à effectuer la<br />
con<strong>de</strong>nsation <strong>de</strong> β-céto-esters et <strong>de</strong> β-dicétone α-allyliques ou α-propoargyliques sur l’aldéhy<strong>de</strong><br />
O<br />

salicylique O-alkylé par <strong>de</strong>s enchaînements insaturés.<br />
Les conditions opératoires que nous avons appliquées, -DBU dans le méthanol-, conduisent à<br />
<strong>de</strong>s ren<strong>de</strong>ments intéressants (qui restent à optimiser), ce qui confirme que le système DBU/ MeOH<br />
est assez indiqué pour ce type <strong>de</strong> con<strong>de</strong>nsation.<br />
L’analyse spectroscopique <strong>de</strong>s produits obtenus a permis d’i<strong>de</strong>ntifier ces différentes<br />
structures qui ont été isolées sous forme d’un seul stéréo-isomère <strong>de</strong> configuration E.<br />
C’est ainsi que quatorze dérivés benzéniques 1,2-polyfonctionnalisés par <strong>de</strong>s groupements<br />
contenant plusieurs liaisons doubles et/ou liaisons triples. D’après nos recherches bibliographiques,<br />
les produits synthétisés ne sont pas décrits dans la littérature.<br />
Par ailleurs, ces structures présentent un potentiel <strong>de</strong> réactivité élevée, du fait <strong>de</strong> la présence <strong>de</strong><br />
plusieurs sites électrophiles et nucléophiles.
Ces composés pourraient donc, éventuellement, être exploités dans différentes réactions comme, par<br />
exemple, <strong>de</strong>s cyclisations donnant <strong>de</strong>s produits polycycliques.

Les solvants ont été distilles sur <strong>de</strong>s <strong>de</strong>sséchants spécifiques sous atmosphère d’argon avant<br />
utilisation.<br />

Chromatographie analytique et préparative :<br />
Les chromatographie sur couche mince (CCM) ont été effectuées sur <strong>de</strong>s plaques <strong>de</strong> silice<br />
«Merck» 60F254 et révélées par le révélateur universel (p-anisaldéhy<strong>de</strong> (5ml), éthanol (90ml),<br />
aci<strong>de</strong> sulfurique (5ml), aci<strong>de</strong> acétique (30gouttes)), l’io<strong>de</strong> ou la lampe Ultra-violet.<br />
Les chromatographies éclair sur gel <strong>de</strong> silice ont été effectuées sur <strong>de</strong> la silice «Merck» 230- 400<br />
Mesh. L’éluant utilise est l’éther éthylique / éther <strong>de</strong> pétrole, dans les proportions (E / Ep).<br />
Données spectroscopiques :<br />
Les spectres ifra-rouge ont été enregistrés sur un spectrophotomètre Perkin-Elmer 1600 à<br />
Transformée <strong>de</strong> Fourier, les fréquences d’absorption sont exprimées en cm -1 .<br />
Les spectres RMN 1 H, 13 C, ont été enregistrés sur appareil « Brucker » AC200 et AC300 MHz et<br />

Mo<strong>de</strong>s opératoires et données spectroscopiques
Procédure générale d’alkylation <strong>de</strong>s β-cétoesters et β-dicétones :<br />
Dans un bicol <strong>de</strong> 250ml sur monte d’un réfrigérant, muni d’une agitation magnétique et place<br />
sous atmosphère d’argon, nous introduisons 40mmol (1eq) <strong>de</strong> β-cétoesters (β-dicétones). Dans<br />
60ml d’acétone anhydre nous ajoutons 160mmol (4 eq) <strong>de</strong> carbonate <strong>de</strong> potassium (K2CO3). Après<br />
15minutes d’agitation à température ambiante nous ajoutons à l’ai<strong>de</strong> d’une ampoule à brome<br />
52mmole (1.3eq) <strong>de</strong> bromure d’allyle (bromure <strong>de</strong> propargyle) dans 60ml d’acétone anhydre. Après<br />
20h au reflux en suivant la réaction par chromatographie sur couche mince (CCM), le milieu<br />
réactionnel est filtré et le solvant est évaporé sous pression réduite .<br />

Dans un bicol <strong>de</strong> 25ml, muni d’une agitation magnétique et sous atmosphère d’argon, on place<br />
le β-cétoesters alkylés ou β- dicétones alkyés, 3mmol (1eq) dans 8ml <strong>de</strong> MeOH anhydre on ajoute<br />
le DBU, 3mmol (1eq) puis le dérivé alkylé <strong>de</strong> l’aldéhy<strong>de</strong> ortho- salicylique, correspondant 3mmol<br />
(1eq) dans 2ml <strong>de</strong> MeOH anhydre à l’ai<strong>de</strong> d’une seringue. On agite à température ambiante,<br />
l’avancement <strong>de</strong> la réaction est suivi par CCM. Le solvant est évapore sous pression réduite Le<br />
résidu huileux est repris avec 20ml d’une solution d’aci<strong>de</strong> chlorhydrique 1N, après extraction avec<br />
<strong>de</strong> l’éther diéthylique (3 × 20ml). Les phases organiques sont ensuite lavées avec une solution<br />
saturée en NaCl (2 × 20ml) et en suite lavées avec <strong>de</strong> l’eau distillée (2 × 20ml) .Après séchage sur<br />

Conclusion générale<br />
Au cours <strong>de</strong> ce travail, nous avons synthétisé une série <strong>de</strong> composés benzénique 1,2<br />
polyfonctionnalisés.<br />
La première partie <strong>de</strong> ce travail a permis <strong>de</strong> mettre au point une stratégie <strong>de</strong> synthèse simple et<br />
économique pour ce type <strong>de</strong> composés.<br />

En effet La con<strong>de</strong>nsation <strong>de</strong>s β-cétoesterts et β-dicétones α-alkylés sur les dérivés O-alkylés <strong>de</strong><br />
l’aldéhy<strong>de</strong> ortho- salicylique, en présence du système -DBU/ MeOH-, a conduit facilement en une<br />
seule étape à <strong>de</strong>s composés benzénique 1, 2 polyfonctionnalisées.<br />
L’application <strong>de</strong> cette stratégie <strong>de</strong> synthèse nous a permis <strong>de</strong> préparer quatorze produits <strong>de</strong> ce<br />
type à partir <strong>de</strong> composés peu coûteux et disponibles (β-cétoesterts , β-dicétones,aldéhy<strong>de</strong>-ortho-<br />
salicylique)<br />
Par ailleurs les structures benzéniques 1,2 polyfonctionnalisées obtenus au cours <strong>de</strong> ce travail<br />
présentent un intérêt synthétique évi<strong>de</strong>nt étant donné la présence <strong>de</strong> divers sites réactionnels<br />
susceptibles <strong>de</strong> réagir.<br />

Parmi Les perspectives <strong>de</strong> ces travaux, l’on peu envisager plusieurs axes <strong>de</strong> développement <strong>de</strong> nos<br />
recherches comme par exemple la cyclisation <strong>de</strong> ces dérivés, par une simple réaction <strong>de</strong> métathèse<br />
pour accé<strong>de</strong>r à <strong>de</strong>s molécules polycycliques.<br />
Nos résultas montrent l'intérêt <strong>de</strong> la stratégie choisie pour la synthèse <strong>de</strong> molécules polycyclique<br />
plus complexes.

Résumé<br />
Les dérivés benzéniques 1,2 polyfonctionnalisés constituent <strong>de</strong>s motifs structuraux importants<br />
en synthèse organique <strong>de</strong> part leur présence dans certaines molécules dont les applications sont<br />
nombreuses et variées (pharmacologiques, biologiques, cosmétiques…… ..).<br />
Au cours <strong>de</strong> ce travail, nous avons synthétisé <strong>de</strong>s dérivés benzénique 1,2 polyfonctionnalisées<br />
à partir <strong>de</strong> réactifs courants et peu coûteux : <strong>de</strong>s β-cétoesters ou β-dicétones, <strong>de</strong> l’aldéhy<strong>de</strong> o-<br />
salicylique et <strong>de</strong>s halogénures d’alkyles.<br />
Les résultas obtenus démontrent l’intérêt <strong>de</strong> la stratégie choisie pour la synthèse <strong>de</strong> ce type<br />
<strong>de</strong> dérivés benzéniques 1,2 polyfonctionnalisés.<br />
L’analyse spectroscopique <strong>de</strong>s composés obtenus a permis d’i<strong>de</strong>ntifier la structure <strong>de</strong>s<br />
différents produits isolés sous la forme d’un seul stéréo-isomère <strong>de</strong> configuration E.<br />
