Deparaf<strong>in</strong>are<br />

Lamele se imerseaza pe rand <strong>in</strong>:<br />

• Xilen 3 x 10 m<strong>in</strong>.<br />

• Etanol 100% 2 x 10 m<strong>in</strong>.<br />

Rehidratarea tesutului<br />

Lamele se imerseaza pe rand <strong>in</strong>:<br />

• Etanol 100% 1 x 10 m<strong>in</strong>.<br />

• Etanol 95% 1 x 10 m<strong>in</strong>.<br />

• Etanol 90% 1 x 10 m<strong>in</strong>.<br />

• Etanol 80% 1 x 10 m<strong>in</strong>.<br />

• Etanol 70% 1 x 10 m<strong>in</strong>.<br />

• Apa distilata 1 x 5 m<strong>in</strong>.<br />

Demascare pentru restaurarea antigenica<br />

Lamele se imerseaza <strong>in</strong>tr-un recipient care cont<strong>in</strong>e tampon citrat pH 6.0 (la temperatura camerei).<br />

Recipientul se s<strong>cu</strong>funda pe jumatate <strong>in</strong> apa d<strong>in</strong> baia termostat aflata la 98ºC unde se <strong>in</strong><strong>cu</strong>beaza 20 de<br />

m<strong>in</strong>ute. Dupa <strong>in</strong><strong>cu</strong>bare se lasa recipientul la racit circa 10 m<strong>in</strong>. la temperatura camerei apoi lamela se<br />

sp<strong>al</strong>a de 2-3 x 5 m<strong>in</strong>. <strong>in</strong> PBS.<br />

Blocarea peroxidazelor endogene<br />

Lamele se imerseaza <strong>in</strong>:<br />

• <strong>1.</strong>5% H2O2 <strong>in</strong> metanol (2,5ml H2O2 30% + 47.5 ml apa distilata) 10 m<strong>in</strong>.<br />

• 1 x 5 m<strong>in</strong>. apa distilata<br />

Blocare biot<strong>in</strong>a endogena<br />

• Imersarea lamelor <strong>in</strong> tampon PBS 1 x 15 m<strong>in</strong>. (tamponul la temperatura camerei)<br />

• In<strong>cu</strong>bare lame <strong>cu</strong> avid<strong>in</strong>a (1mg/ml) 1 x 20 m<strong>in</strong>.; lama s<strong>cu</strong>rsa b<strong>in</strong>e <strong>si</strong> <strong>in</strong><strong>cu</strong>bata <strong>cu</strong> 50-100 µl de solutie<br />

• Sp<strong>al</strong>are <strong>in</strong> PBS rapida pentru <strong>in</strong>laturarea grosului de solutie de avid<strong>in</strong>a<br />

• In<strong>cu</strong>bare <strong>in</strong> PBS 1 x 5m<strong>in</strong>.<br />

• In<strong>cu</strong>bare lame <strong>cu</strong> 100 µl biot<strong>in</strong>a (0.1mg/ml) 1 x 20 m<strong>in</strong>.<br />

• Sp<strong>al</strong>are <strong>in</strong> PBS rapida pentru <strong>in</strong>laturarea grosului de solutie de biot<strong>in</strong>a<br />

• In<strong>cu</strong>bare <strong>in</strong> PBS 3 x 5m<strong>in</strong>.
