C. Other Kinase Assay Formats (non-radioactive)<br />

The PepTag® Protein Kinase Assays are fast <strong>and</strong><br />

quantitative non-radioactive alternatives to<br />

[γ-32P]ATP-based assays for measuring protein kinase C<br />

(Cat.# V5330) <strong>and</strong> cAMP-dependent protein kinase (Cat.#<br />

V5340) activity. The assays use fluorescently-tagged peptide<br />

substrates with a net positive charge. Phosphorylation<br />

changes the charge of the peptide to a net negative, which<br />

influences the migration of the peptide in an agarose gel.<br />

This is the basis for detecting changes in phosphorylation<br />

via a rapid, 15-minute agarose gel separation (Figure 7.12).<br />
