1) What is the growth characteristics <strong>of</strong> the recombinant yeast? 1s the <strong>production</strong> <strong>of</strong> the<br />

recombinant <strong>protein</strong> growth-associated?<br />

2) What is the nature <strong>of</strong> the plasmid instability in the recombinant yeast? Are the 2<br />

based yeast shunle piasmids stable? And if not, to what emt?<br />

3) How do fermentation conditions, growth rates and growth media affect the plasaid<br />

stability and the recombinant <strong>protein</strong> <strong>production</strong>?<br />

4) How are the metaboüc pathways in the yeast related to the recombinant <strong>protein</strong><br />

formation.<br />

5) What is the advantages and disadvantages <strong>of</strong> asing an airlift <strong>bioreactor</strong> for the<br />

recombinant yeast fementatior<br />

4 3 Batch Suspension Fermentation<br />

Batch suspension cultrms were carried out in a 15 Iitu airiift bionxctor (1.2 iitet working<br />

volume) at 30 O C and an aeration rate <strong>of</strong> 1.0 VVM. Figure 4.1 shows a ~ypical time<br />
