5,2,1,3 Antibiotics<br />
Because <strong>of</strong> expected widespread penicill<strong>in</strong>/streptomyc<strong>in</strong> resistance among local<br />
bacterial stra<strong>in</strong>s, various antimicrobial agents (such as Gentamyc<strong>in</strong>, Vancomyc<strong>in</strong>,<br />
<strong>and</strong> low dose Amikac<strong>in</strong>) were used over <strong>and</strong> above the penicill<strong>in</strong>/streptomyc<strong>in</strong><br />
orig<strong>in</strong>ally <strong>in</strong>corporated <strong>in</strong>to the <strong>culture</strong> medium.<br />
While the optimisation experiments were undertaken, an aerophilic laboratory<br />
contam<strong>in</strong>ant, Xanthomonas maltophilia (identified by biochemical reactions <strong>in</strong> the<br />
Api 20E system (BioMerieux) was detected on some <strong>culture</strong> tubes <strong>of</strong> the WB<br />
reference stra<strong>in</strong> (the trophozoites cont<strong>in</strong>ued to grow <strong>and</strong> multiply <strong>in</strong> the presence<br />
<strong>of</strong> these microbes). A disk-diffusion susceptibility assay revealed that the<br />
organism was resistant to the antibiotics <strong>in</strong>corporated <strong>in</strong> the medium, <strong>in</strong>clud<strong>in</strong>g<br />
Amikac<strong>in</strong> but sensitive to Cipr<strong>of</strong>loxac<strong>in</strong>. Subsequently, this antibiotic was required<br />
to elim<strong>in</strong>ate the bacterial contam<strong>in</strong>ants without <strong>in</strong>hibit<strong>in</strong>g the growth <strong>of</strong> Giardia<br />
trophozoites. An assay to determ<strong>in</strong>e the Cipr<strong>of</strong>loxac<strong>in</strong> concentration tolerated by<br />
trophozoites was therefore performed, as literature cit<strong>in</strong>g use <strong>of</strong> this quionolone on<br />
Giardia <strong>culture</strong>s was not found.<br />
