6.3 RESULTS<br />

Trophozoites have been successfully preserved for periods vary<strong>in</strong>g from 23 days<br />

to approximately 2 years. Qualitative assessment upon retrieval revealed 70-80%<br />

viability <strong>of</strong> trophozoites. With<strong>in</strong> 30 m<strong>in</strong>utes, more than 50% <strong>of</strong> the trophozoites<br />

were adher<strong>in</strong>g to the <strong>in</strong>ner surface <strong>of</strong> the glass <strong>culture</strong> tubes (Plate 6.1). Confluent<br />

cells were seen with<strong>in</strong> 48h <strong>of</strong> <strong>culture</strong> (Plates 6.2 & 6.3). All retrieved samples<br />

have been re-established <strong>in</strong> <strong>culture</strong>: a 100% success rate has been achieved<br />

us<strong>in</strong>g both electronically controlled (6 times) <strong>and</strong> mechanical (15 times) cool<strong>in</strong>g<br />

techniques. Table 6.1 presents the records <strong>of</strong> duration <strong>of</strong> preservation <strong>of</strong> the 6<br />

local isolates <strong>and</strong> the two reference stra<strong>in</strong>s.<br />

Viable <strong>culture</strong>s <strong>of</strong> the retrieved trophozoites have been ma<strong>in</strong>ta<strong>in</strong>ed <strong>in</strong> <strong>culture</strong> for<br />

several months <strong>and</strong> cryopreserved aga<strong>in</strong> with success. A reserve supply<br />

(cryobank) has been established for the 6-axenised local stra<strong>in</strong>s.<br />
