5.3.3 In <strong>vitro</strong> <strong>culture</strong> <strong>of</strong> <strong>in</strong> vivo-derived trophozoites<br />

Fifty-three mice litters were <strong>in</strong>oculated with cysts for <strong>in</strong> vivo excystment <strong>and</strong><br />

subsequent trophozoite cultivation <strong>in</strong> <strong>vitro</strong>. Trophozoites were harvested from 20<br />

<strong>of</strong> those <strong>and</strong> <strong>in</strong>oculated <strong>in</strong>to TYI-S-33 medium (the results are summarised <strong>in</strong><br />

Table 5.4). Of the 20 attempted isolations, 14 failed to establish viable <strong>culture</strong>s.<br />

Eight <strong>of</strong> the 14 isolates died even though they were not contam<strong>in</strong>ated; they<br />

survived from 12 hours to seven days <strong>in</strong> <strong>culture</strong>, but eventually rounded up <strong>and</strong><br />

died. Six were overwhelmed by contam<strong>in</strong>ants. Six isolates were successfully<br />

adapted to <strong>culture</strong> <strong>and</strong> subsequently, the first axenic South African stra<strong>in</strong>s were<br />

established <strong>in</strong> <strong>vitro</strong>. They have been ma<strong>in</strong>ta<strong>in</strong>ed <strong>in</strong> <strong>culture</strong> for over 15 months.<br />

Stabilates <strong>and</strong> Iysates <strong>of</strong> the axenised stra<strong>in</strong>s were cryopreserved for future use.<br />

They have been arbitrarily assigned numbers correspond<strong>in</strong>g with the<br />

sample number, preceded by the prefix SA (for South Africa) viz SA6, SA7, SA18,<br />
