A broth concentrate (five times concentrated by <strong>in</strong>creas<strong>in</strong>g the concentration <strong>of</strong> the<br />

<strong>in</strong>gredients listed on p182 five times) conta<strong>in</strong><strong>in</strong>g Biosate, glucose <strong>and</strong> sodium<br />

chloride was prepared <strong>and</strong> filter sterilised. This medium can be stored at 4°C for<br />

up to 1 year. A buffer conta<strong>in</strong><strong>in</strong>g 5x the concentration <strong>of</strong> the buffer salts was also<br />

made. A 6,5% bile solution <strong>and</strong> 2,28mg/ml Ferric ammonium citrate were<br />

prepared separately. All stock solutions were stored at 4°C. The complete medium<br />

was made by dilut<strong>in</strong>g the broth concentrate to 500 millilitres <strong>of</strong> distilled water, 10%<br />

serum, antibiotics, <strong>and</strong> a 5x concentrated, freshly prepared reduc<strong>in</strong>g solution <strong>of</strong> L­<br />

cyste<strong>in</strong>e <strong>and</strong> Ascorbic acid. The complete medium was used with<strong>in</strong> 1 week <strong>of</strong><br />

preparation.<br />

Our modification <strong>in</strong>volved use <strong>of</strong> the other am<strong>in</strong>oglycosides because <strong>of</strong> expected<br />

widespread penicill<strong>in</strong>/streptomyc<strong>in</strong> resistance among local bacterial stra<strong>in</strong>s.<br />
