Organisations: • Applied and Analytical Chemistry • Institute for Materials Research Researchers: • Robert CARLEER Bilateral co-operation: Determination of toxic compounds and metal's speciation in fish in industrial areas Universiteit Hasselt Abstract: The aim of the study is to determine toxic compounds and speciation of metals in freshwater fish collected from several industrial and agricultural areas in Dolnysk in Poland. The evolution of pollutants profiles as a function of the following factors such as location, season, fish organ, fish species will be studied. The relationship between the kind of industrial activity and the kind, form and distribution of pollutants in fish will be evaluated. Fish absorb the pollutants by skin, gills and with food; thus, its health depends on the quality and conditions of water. Often, fish tissues bioaccumulate and thus contain higher level of concentration of pollutants than water they live in. The main exposure route to contaminants by life organisms is food. One of humans diet parts are fishes which are able to accumulate in their organisms a lot of toxic substances. Therefore, it is important to monitor the contaminants of inorganic and organic origin in fish tissues. The work will also be focused on persistent organic pollutants (POPs), including e.g. polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), dioxins, furans and organochlorine pesticides. These compounds are highly toxic, including for humans and other living organisms that share the environment. Once released into the environment they may be transported over long distances and pass trough many biochemical cycles, e.g. food web, without undergoing significant degradation. A very important part for this work is the collection, storage and preparation of fish samples for analysis from which the result is strongly dependent on the procedures applied in this step. The main topic is the elaboration of methods for tissue sample preparation to eliminate fatty acids and lipids fraction which preclude the identification of some organic compounds due to a strong interference. There are several methods of cleaning-up which will be tested and optimized such as adsorption using different column packing (Al2O3, CaO, florisil) and freezing-lipid extraction technique. The second step will be concerned with the elaboration of effective method for isolation of POPs. Soxhlet extraction, ultrasonication, microwave-assisted extraction and other techniques using different organic solvents and their mixtures will be considered and optimized. The fractionation of the obtained extract by adsorption chromatography will be developed. Taking into account that fish tissue samples contain a large quantity of water, therefore, a step of lyophilization should also be included. The most important analytical techniques, which are planned to be applied for the determination of POPs and their concentration, are the following: gas chromatography with different detection systems (GC-MS, GC-FID, GC-ECD) and high-performance liquid chromatography (HPLC). It is known that the simultaneous analysis of different POP groups generates a lot of difficulties. Thus, the development of integrated methods will also be undertaken. Most of trace metals and metalloids play a significant role in the functioning of living organisms, whereas there are some that can be highly toxic to animals and humans. To study the level of contamination of fish tissues with metals different spectroscopic techniques will be used such as atomic absorption spectroscopy (AAS) and with inductive coupled plasma atomic emission spectroscopy (ICP-AES). I would like to monitor selected heavy metals (Pb, Hg, As, Cd, etc.) and trace elements that play fundamental role in the proper functioning of organism which are e.g. Fe, Zn, Cu or Se and that could be toxic in high doses. The use of HPLC coupled with ICP-MS to analyze inorganic and organic species of selected elements is considered if the access to this technique will be reached. Sample preparing is a crucial step in trace and speciation of metals analysis. Besides a lot of metals change their speciation forms into another very rapidly. The main factors in getting reliable results are the time delay from fish collection to analysis and preservation of samples. The most popular method of fish tissues preservation is cooling/freezing. Moreover, it is very difficult to analyze different speciation of some elements simultaneously; nevertheless it is a challenge to develop a method for analyzing different metal-organic speciation of one element at one step. In my work I would like to make an attempt to develop and optimize the method of preparing samples and match the most suitable instrumental technique of analysis. The application of HPLC coupled with ICP-MS seems to be very promising for analysis of organic species of As, Se and Cd. The determination of metalloids species like metalloproteins, metallosugars, methallotioneins etc. will be considered. For this purpose gel permeation chromatography (GPC) or size exclusion chromatography (SEC) coupled with ICP-MS should be useful in the separation of different metallobiomolecules and analysis of metalloorganic species. There must be taken into consideration an aspect of interferences during spectral analysis coming because of the complexity of the biological matrix. A particular attention will be paid on minimizing influences coming from the background and spectral interferences connected with particular elements and mutual interactions.
