IMPORTANCE, AIMS AND POSSIBLE OUTCOME OF RESEARCH<br />
Sepsis is one of the biggest challenges in medicine because of its rapidly proliferating<br />
nature. On account of the complexity of its pathomechanism, sepsis is hard to diagnose,<br />
and amongst the countless therapeutic possibilities used, there is no perfect solution yet.<br />
A common feature of septic multiorgan failure (MOF) is microcirculatory dysfunction,<br />
which leads to tissue hypoperfusion, mitochondrial dysfunction and necrosis. Thus,<br />
microcirculatory-mitochondrial resuscitation seems to be a promising therapeutic target.<br />
In our previous studies we have shown that the selective modulation of the endothelin-1<br />
(ET-1) receptors – the most potent vasoactive agent of the body – can be a therapeutic<br />
solution for sepsis on macrohemodynamics, microcirculation and mitochondrial<br />
respiration too. Nevertheless, it is unknown if the proven mechanism is a direct or indirect<br />
effect on mitochondria and if it has any advantages compared to the today used inotropic<br />
therapies.<br />
AMBITIONS AND CAREER GOALS<br />
In this project my first goal is to learn in vivo techniques which I can perform on rodents<br />
and minipigs. I am interested in the technique of high-resolution respirometry (Oroboros<br />
O2k), which is a useful tool to investigate mitochondrial functions. I would like to expand<br />
both my theoretical and practical knowledge, which I will be able to use either in medical<br />
research or in medical practice.<br />
