4.2.3 E'xperimenlal Prolm-oh<br />

Initially, a dose-response<br />

analysis was undertaken with various doses for each <strong>of</strong><strong>the</strong> live<br />

different drugs used in <strong>the</strong> experimental protocols. The drugs were applied to <strong>the</strong><br />

sections for 10min and <strong>the</strong>n washed out after 30min <strong>of</strong> stabilisation, followed by<br />

prolonged ischaernia and reoxygenation. Once <strong>the</strong> optimal dose for each drug was<br />

determined, <strong>the</strong> preparations were randomly allocated to various protocols (n-6/group).<br />

In <strong>the</strong> groups subjected to simulated ischemia, this was induced for a period <strong>of</strong> 90min and<br />

<strong>the</strong>n it was followed by 120min <strong>of</strong> reperfusion. The time course and <strong>the</strong> incubation times<br />

with <strong>the</strong> various agents are shown in Figure 4.1<br />

4.2.4 Assessment <strong>of</strong> tissue injury and viability:<br />

Tissue injury was analysed as described in section 3.2.4 (Chapter 3).<br />
