1.9 Aims <strong>of</strong> <strong>the</strong> <strong>the</strong>sis<br />

The aims <strong>of</strong> <strong>the</strong> <strong>the</strong>sis are to investigate <strong>the</strong> optimal <strong>preconditioning</strong> protocol in <strong>the</strong><br />

<strong>human</strong> myocardium and <strong>the</strong> specific role played by K.<br />

x-1-1. channels as an effector <strong>of</strong><br />

<strong>ischaemic</strong> <strong>preconditioning</strong> protection. These experiments are based on an in 01ro model<br />

<strong>of</strong> <strong>ischaemic</strong> <strong>preconditioning</strong> using <strong>human</strong> right atnal appendage myocardium.<br />

1. To charactense <strong>the</strong> short and prolonged effects <strong>of</strong> ischaernia and reoxygenation in this<br />

modcl.<br />

1. To characterise <strong>the</strong> <strong>ischaemic</strong> <strong>preconditioning</strong> phenomenon in <strong>the</strong> <strong>human</strong><br />

myocardium.<br />

3. To investigate <strong>the</strong> role <strong>of</strong> KATP channels as an effector in <strong>the</strong> protection induced by<br />

<strong>ischaemic</strong> <strong>preconditioning</strong> in <strong>the</strong> <strong>human</strong> myocardiurn<br />

4. To investigate <strong>the</strong> effects <strong>of</strong> age and common pathological conditions i. e. diabetes<br />

mellitus, poor cardiac function on <strong>the</strong> protection induced by <strong>ischaemic</strong><br />

<strong>preconditioning</strong>.<br />

5. To investigate <strong>the</strong> protection <strong>of</strong> <strong>the</strong> <strong>human</strong> <strong>heart</strong> against myocardial tissue damage by<br />

<strong>ischaemic</strong> <strong>preconditioning</strong> in an itt Wvo clinical setting.<br />
