2.12 Conclusiow<br />

I have characterised a model <strong>of</strong> ischaernia and reoxygenation <strong>of</strong> <strong>human</strong> myocardium<br />

using right atrium appendage obtained from patients undergoing cardiac surgery.<br />

The tissue is readily available and <strong>the</strong> preparation is inexpensive. The preparation is<br />

stable for at least 24 hr and this permits <strong>the</strong> study <strong>of</strong> <strong>the</strong> early and delayed<br />

consequences <strong>of</strong> ischaemia and reperfusion. In addition, <strong>the</strong> extended stabilltv <strong>of</strong> <strong>the</strong><br />

model may be potentially used for genetic manipulation to investigate <strong>the</strong><br />

pathophysiological mechanisms underlying <strong>the</strong> injury sustained during Ischaemia<br />

and reperfusion, and to develop new <strong>the</strong>rapeutical strategies to combat <strong>the</strong>ir<br />

undesirable effects. Once, <strong>the</strong> model was characterised, I will be able to use it to<br />

investigate <strong>the</strong> phenomenon <strong>of</strong> ischernic <strong>preconditioning</strong> in <strong>the</strong> <strong>human</strong> myocardium.<br />

The charactefisation <strong>of</strong> <strong>the</strong> early and delayed windows <strong>of</strong> protection has not been<br />

explored in full and is <strong>the</strong> subject <strong>of</strong> <strong>the</strong> third chapter<br />
