again by 48 hr oxygen consumption dramatically decreased to 20% <strong>of</strong> <strong>the</strong> initial<br />

values (184 1 12 nmol 02/g wet wt/min, p-, 0.05). These findings fur<strong>the</strong>r support<br />

that this <strong>human</strong> atfium preparation is viable and stable for at least 24 hr <strong>of</strong><br />

incubation but not for 48 hr.<br />

phosphate content <strong>of</strong> <strong>the</strong> slices following <strong>the</strong> various time periods <strong>of</strong> aerobic<br />

incubation. The ATP, ADP and AMP contents, ADP/ATP ratio and <strong>the</strong> total<br />

adenylate pool were all similar in <strong>the</strong> slices aerobically incubated for 0.5,4 and 24<br />

hr. However, in line with previous results <strong>the</strong>re was a significant reduction in all<br />

metabolites after 48 hr <strong>of</strong> aerobic incubation.<br />

Morphological Assessment<br />

The results <strong>of</strong> <strong>the</strong> semi-quantative morphological examination are shown in Figures<br />

2.9. Figures 2.1 Oa, b and c show representative high resolution electron transmission<br />

micrographs <strong>of</strong> grade I (normal), grade 2 (mild damage) and grade 3 (severe<br />

damage). The morphological appearance <strong>of</strong> specimens aerobically perfused for 0.5,<br />

4 and 24 hr was similar, however, significant damage was observed in specimens<br />

aerobically perfused for 48 hr.<br />
