Le GPR91 et la préservation <strong>de</strong>s neuronesL’étu<strong>de</strong> réalisée par l’équipe est éga<strong>le</strong>ment prometteuse quant à lapossibilité que <strong>le</strong> GPR91 préserve <strong>le</strong>s neurones. « Les neurones sont<strong>le</strong>s principaux senseurs participant à l’oxygénation <strong>de</strong> la rétine et <strong>le</strong>sprincipaux intervenants dans <strong>le</strong> processus <strong>de</strong> réparation <strong>de</strong> cel<strong>le</strong>-ci »,explique <strong>le</strong> Dr Sylvain Chemtob, directeur <strong>de</strong> l’étu<strong>de</strong>, chercheur dans<strong>le</strong> domaine néonatal au CHU Sainte-Justine et professeur audépartement <strong>de</strong> pédiatrie, d’ophtalmologie, <strong>de</strong> pharmacologie et àl’éco<strong>le</strong> d’optométrie <strong>de</strong> l’Université <strong>de</strong> Montréal.« Compte tenu <strong>de</strong>s similarités entre la rétine et <strong>le</strong> cerveau, nosconclusions au sujet <strong>de</strong> la rétine pourront être éga<strong>le</strong>ment appliquées aucerveau, affirme <strong>le</strong> Dr Chemtob. En stimulant <strong>le</strong> développement duréseau vasculaire l’activation du récepteur GPR91 pourrait éga<strong>le</strong>mentpermettre <strong>de</strong> préserver <strong>le</strong>s neurones <strong>de</strong>s tissus cérébraux endommagéslorsqu’une personne subit un acci<strong>de</strong>nt vasculaire cérébral ou untraumatisme crânien. »Sylvain ChemtobLe GPR91 pour stopper <strong>le</strong>s tumeursL’étu<strong>de</strong> en question est la toute première à porter <strong>sur</strong> <strong>le</strong>s vastes répercussions <strong>de</strong> l’utilisation duGPR91 et la manière dont ce récepteur, présent <strong>sur</strong> <strong>le</strong>s neurones, réagit aux stress et ajuste l’étatd’oxygénation <strong>de</strong>s tissus.« Il s’agit d’un nouveau concept en biologie vasculaire, souligne <strong>le</strong> Dr Sapieha, qui note qu’on peutenvisager <strong>de</strong> lutter contre l’expansion <strong>de</strong>s tumeurs cancéreuses en travaillant avec <strong>le</strong> récepteurGPR91. Si, en bloquant <strong>le</strong> récepteur GPR91, on empêche <strong>le</strong>s vaisseaux sanguins <strong>de</strong> proliférer etd’alimenter une tumeur en nutriments et en oxygène, on pourra entraver considérab<strong>le</strong>ment laprogression du cancer. »Même si cette <strong>recherche</strong> prometteuse a été menée <strong>sur</strong> <strong>de</strong>s animaux, <strong>le</strong> récepteur GPR91 estéga<strong>le</strong>ment présent chez <strong>le</strong>s humains. Volume 6, Numéro 5 • Juin 2009Recherche et publicationsAdriana Di Polo participe à une découverte majeure pour <strong>le</strong>smaladies neurologiquesAdriana Di Polo, du département <strong>de</strong> pathologie et biologiecellulaire, en compagnie <strong>de</strong> chercheurs <strong>de</strong> l'U<strong>de</strong>M et <strong>de</strong>l'Université McGill, a découvert que <strong>de</strong>s cellu<strong>le</strong>s quisoutiennent habituel<strong>le</strong>ment la <strong>sur</strong>vie <strong>de</strong> cellu<strong>le</strong>s nerveuses(neurones) jouent aussi un rô<strong>le</strong> actif et majeur dans la mort <strong>de</strong>neurones dans l'œil. Les résultats <strong>de</strong> <strong>le</strong>urs travaux, publiés dansThe Journal of Neuroscience, pourraient ouvrir la voie à <strong>de</strong>sthérapies plus efficaces pour divers troub<strong>le</strong>s neurologiques aigus et chroniques, dont<strong>le</strong> glaucome et l'occlusion <strong>de</strong> l'artère centra<strong>le</strong> <strong>de</strong> la rétine.Selon Adriana Di Polo : « L'aspect <strong>le</strong> plus intéressant <strong>de</strong> notre étu<strong>de</strong>, et la raison pourlaquel<strong>le</strong> nous sommes si enthousiastes, est que la voie qui mène à la mort déc<strong>le</strong>nchéepar <strong>le</strong> glutamate <strong>de</strong>s cellu<strong>le</strong>s nerveuses met en cause un autre joueur vital – à savoir,<strong>le</strong>s cellu<strong>le</strong>s glia<strong>le</strong>s.
