[teleomorphGlomerella cingulata (Stoneman) Spauld.&Schrenk.] causes brown blight of tea [Camellia sinensis(L.) O. Kuntze] plant, which is a woody perennial crop. Differentvarieties are expected to vary in tolerance levels to aparticular pathogen. Since proteins are important componentsof any plant, which reflect the way in which the plantreacts to the pathological and other stresses, it was decidedto conduct total soluble protein profile analysis of healthyand pathogen inoculated tea leaves of different varieties.Eighteen tea varieties obtained from Tea GermplasmBank, Botany Department, were initially screened for resistanceagainst G. cingulata following the detached leaf inoculationmethod (Dickens and Cook,1989), percentage lesionproduced being inversely proportional to the resistance.Total soluble proteins of tea leaves of different varietieswere extracted (Chakraborty et al, 1995), estimated (Lowryet al, 1951) and analysed by SDS-PAGE (Sambrook et al,1989) according to the standard procedures.It is evident from the table that in the susceptible varieties(TV-22, T-17, UP-26, CP-1/1, BSS-3) the protein contentdecreased significantly. SDS- PAGE analysis of theseextracted proteins reflected the protein content changes,with decreased content showing reduction in number ofbands. On the other hand, in the resistant varieties (TV-18,TV-30, BSS-2, UP-9, P-312, TV-25 and TV-26) there wasno significant change in total protein content on infection.In these varieties leaf proteins of ca. 97.4, 94.3 kDa weredegraded by the fungus. New proteins were not induced. Insome of these varieties (TV-30, P-312, TV-25 and TV-36)these two proteins were newly synthesized on inoculationwith G. cingulata. Thus, these seem to be associated withresistance. Surprisingly, their initial presence in the host isnot associated with resistance. It is interesting that in somemoderately resistant varieties (TV-29, BSS-2, BS/7A/76and AV-2) the banding pattern did not change at all.Therefore, total protein analysis throws some light onthe particular proteins involved in the resistance towardsbrown blight pathogen. It is also clear that the fungus degradessome proteins of the host, breaking down its resistancemechanism, while the resistant varieties were able to tacklethe stress by maintaining the pool of the total proteins.1.
